Rape Pollen Supercritical Co <sub> 2 </ Sub> Extracts On The Immune And Inflammatory

At first,this study aimed to evaluate the effect of extract bysupercritical carbon dioxide extraction from rape pollen on mice immunefunctions,including some indexes about inflammation in chief. The micewere randomized into four groups including normal control group,lo-pollen's extraction group, mid-pollen's extraction group, andhi-pollen's extraction group. After three weeks of perfusion diets intostomach, the mice were executed, and thymus index,spleen index,Tlymphocyte proliferation index and B lymphocyte proliferation indexwere determinated. The concentration of IL-2,IL-4,IFN-γ in plasma andin T cells were detected. Also the concentration IL-1βin plasma and inmacrophages were detected.Thymus index and spleen index were lower in the pollen's extractiongroups than NC group. The proliferation indexes both of T and Blymphocytes were decreased in mice fed pollen's extraction. The plasmalevels of IL-2,IFN-γ and IL-1β in all pollen's group were significantlylower than NC group, especially in HPE group and MPE group. But IL-4has changed little. Lymphocytes secretion levels of IL-2,IFN-γ andIL-1β were decreased in mice fed pollen's extraction. But IL-4 has nochang,too. Then we examined potential therapeutic anti-inflammatory effectsof extract by supercritical carbon dioxide extraction from rape pollen.Rheumatoid arthritis is one of autoimmue diseases, belonging to chronicinflammation disease. The RA mice model were established in ourtrials.The mice were randomized into three groups including normalcontrol group, CIA model control group, mid-pollen's extraction group.The onset of paw swelling was on day 21 after injection of chickencollagentype II and the swelling peak appeared on day 28-36 and thendeclined after day 40. Spleens index were assayed on day 44 after im-munization. The results showed that spleen indices of CIA mice wereincreased. Pollen's extraction reduced the elevated spleens index of CIAmice. T and B cells proliferation in CIA mice were increased andpollen's extraction could suppress the elevated proliferation of T cellsand B cells stimulated by ConA and LPS respectively. The production ofIL-lβ by macrophages and IL-2, IFN-γ by T cells were upregulatedsignificantly in CIA model. Pollen's extraction decreased the productionof IL-1β, IL-2 and IFN-γ. But IL-4 secretion level has no change inserum and T cells.All these results suggested that pollen extract which contain themainly component of ALA had antiinflammation and immuemodulatoryeffects.
