In this article, HPLC/MS is used to analyze the nucleotides in the fruit body of the cultured Cordyceps kushuensis Kob. In the experiment, firstly we optimize the chromatography conditions. Then the quality and quantity of the nucleotides are accurately determined according to the following gradient program:0-5min: from99%H2O+1%ACE to 96%H2O+4%ACE, 5-30min: 96%H2O+4%ACE. The results show that there are five kinds of nucleotides in the fruit body of the cultured Cordyceps kushuensis Kob and the content of cordycepin in the fruit body of the cultured Cordyceps kushuensis Kob is 4.356mg/g, which is more than that of cordycepin in the fruit body of Cordyceps sinensis.. So the cultured Cordyceps kushuensis Kob can be considered as the main source of cordycepin.In the early period of studying cordycepin clinically, people found that the enzyme can degenerate cordycepin easily so that cordyceptn will decrease the effect of medicine before it works in the body. So it is necessary for us to do the research about how to improve the medical potential, decrease the using content and cost of cordycepin. With the wide use of the technology of nanohybnde, researchers are trying to combine this technology with the research of the herbal medicine. We try to apply the technology of nanohybride to the study of cordycepin, so in this article we use the unstable coprecipitation method to prepare the layered double hydroxide (LDH), then isolate and purify the cordycepin from the cultured Cordyceps kushuensis Kob. After that cordycepin/LDH nanohybride is successfully synthesized by direct interaction of cordycepin into LDH. The resulting nanohibride are characterized using XRD, IR, CZE and TEM. At last we primarily study the anti-tumor activities in vitro. The results show that the co'rdycepin/LDH nanohybride can distinctly inhibit the growth of the U937.Meanwhile the supercritical fluid carbon dioxide extraction technology together with other methods is used to isolate and purify the flavones in the cultured Cordyceps kushuensis Kob.. In this experiment we use the single factor experiment and the orthogonal experiment to explore the prime craftwork and compare the recoveries and varieties of the flavones extracted by different ways.
