The SSR reaction system of peach was optimized: total volumewas 25μL, including 80 ng template DNA,1×PCR Buffer,2 U Taq DNA polymerase,MgCl₂ 2 mmol/L,dNTPs 240-360μmol/L,0.4μmol/L each of specific primers, the reactionis: 94℃5 mins; 94℃1 min,53℃30s,72℃1 min, 35cycles; 72℃5 mins.There are only UDP96-001 and UDP96-416 which amplify 3 markers have linkage withpollen sterility gene in the total 9 pairs of primer. The genetic distance is 12cM to Ps/psgene. Other two locus(UDP96-460bp,95bp) of genetic distance are 5 cM,8 cM,to the Ps/psgene, because the two marker are between the Ps/ps, and the distance is 13cM,so it can beused in MAS when both be used together.The ps gene is on the six linkage group through theexperiment,it may gives rule for the fine mapping.
