Research Of The Effect Of Pqs On The Expression Of Caspase-8 And Caspase-9 On Cerebral Ischemia

ObjectiveTo investigate the effects and the further mechanism of Panax quinquefolium saponins from steams and leaves (PQS) on Caspase-8 and Caspase-9. MethodsIn order to observe the neuroprotective effect of PQS on cerebral ischemia, the rats with incomplete cerebral ischemia were induced with fishing lines in the middle cerebral artery (MCA).The following indexes were determined:①The neurological deficit and pathological change were deterinimed by Longa's score, optical microscope;②Caspase-8,Caspase-9 mRNA in the cortical neuron were detected with reverse transcription polymerase chain reaction (RT-PCR).Results1.PQS significantly ameliorated the neurological deficit and improved the pathological changes.2. PQS significantly decreased the apoptosis of neural cells, reduced the expression of Caspase-8 and Caspase-9 mRNA . ConclusionPQS had protecting effect on cerebral ischemia,the mechanism of PQS against cerebral ischemia is related to inhibiting cell apoptosis.
