Objective:To investigate expression and significance of PI3K, mTOR, Pokemon, and Ku70 in nasopharyngeal carcinoma. Methods:Immunohistochemical Envision method was adopted to determine the expression of PI3K, mTOR, Pokemon and Ku70 in 47 cases of nasopharyngeal carcinoma and 20 cases of nasal polyp, and to retrospectively analyze their correlations to the clinicopathologic characteristics of NPC. Results:1) Positive expression rate of PI3K in NPC group and nasal polyp group is 76.6%(36/47) and 35.0%(7/20) respectively. There is a statistics difference between two groups (χ2=12.36, P<0.05). While in different pathological types of NPC tissues, positive expression rate of PI3K in groups of keratinizing squamous cell carcinoma and non-keratinizing carcinoma is 65.0% and 85.1% respectively, and the expression difference shows no statistical significance (χ2=2.61, P<0.05). The positive expression indicates that PI3K is related to occurrence of tumor, but not closely related to tumor types. In different TNM NPC tissues,Ⅰstage+Ⅱstage group,Ⅲstage+Ⅳstage group, positive expression rate of PI3K is 80.0% and 72.7% respectively, and the difference shows no statistic significance (χ2=0.35, P>0.05), which indicates that PI3K is not closely related to neoplasm staging. Positive expression rate of PI3K in NPC tissues with or without lymphatic metastasis is 70% and 88.2% respectively, and the difference shows no statistic significance (χ2=2.91,P>0.05), which indicates that PI3K is not closely related to lymphatic metastasis; 2) Positive expression rate of mTOR is in NPC group and nasal polyp group is 70.2% (33/47) and 25.0%(5/20) respectively, and the difference is of statistical significance (χ2=11.6, P<0.05). In different types of NPC issues, positive expression rate of mTOR in groups of keratinizing squamous cell carcinoma and non-keratinizing carcinoma is 55.0% and 81.5% respectively, and the expression difference is of statistical significance (χ2=3.85, P<0.05), which shows that mTOR is related to occurrence of tumor, and is closely related to pathological types. In different TNM NPC tissues,Ⅰstage+Ⅱstage group,Ⅲstage+Ⅳstage group, mTOR positive expression rate is 72.0% and 68.2% respectively, and the difference shows no statistic significance (χ2=0.08, P>0.05), which indicates that mTOR is not closely related to neoplasm staging. Positive expression rate of mTOR in NPC tissues with or without lymphatic metastasis is 76.7% and 58.88% respectively, and the difference shows no statistic significance (χ2=1.65, P>0.05), which indicates that mTOR is not closely related to lymphatic metastasis; 3) Positive expression rate of Pokemon in NPC group and nasal polyp group is 68.1%(32/47) and 25.0%(5/20) respectively, and the difference is of statistical significance (χ2=10.53, P<0.05). In different types of NPC issues, positive expression rate of Pokemon in groups of keratinizing squamous cell carcinoma and non-keratinizing carcinoma is 45.0% and 85.2% respectively, and the expression difference is of statistical significance (χ2=8.54, P<0.05). The positive expression shows that Pokemon is related to occurrence of tumor, and is closely related to pathological types. In different TNM NPC tissues,Ⅰstage+Ⅱstage group, andⅢstage +IVstage group, positive expression rate of Pokemon is 88.0% and 45.2% respectively, and the difference is of statistic significance (χ2=9.75, P<0.05), which indicates that Pokemon is closely related to neoplasm staging. The positive expression rate of Pokemon in NPC tissues with or without lymphatic metastasis is 86.7% and 35.5% respectively, and the difference is of statistic significance (χ2=13.18, P<0.05), which indicates that Pokemon is closely related to lymphatic metastasis. (4) Positive expression rate of Ku70 in NPC group and nasal polyp group is 61.7%(29/47)and 30.0%(6/20) respectively, and the difference is of statistical significance (χ2=5.65, P<0.05). In different types of NPC issues, positive expression rate of Ku70 in groups of keratinizing squamous cell carcinoma and non-keratinizing carcinoma is 55.0% and 66.7% respectively, and the expression difference shows no statistical significance (χ2=0.66, P>0.05). The positive expression shows that Ku70 is related to tumor, but is not closely related to pathological types. In different TNM NPC tissues,Ⅰstage+Ⅱstage group, andⅢstage+Ⅳstage group, positive expression rate of Ku70 is 72.0% and 50.0% respectively, and the difference shows no statistic significance (χ2=2.40, P>0.05), which indicates that Ku70 is not closely related to neoplasm staging. The positive expression rate of Ku70 in NPC tissues with or without lymphatic metastasis is 73.3% and 35.5% respectively, and the difference is of statistic significance (χ2=4.75, P<0.05), which indicates that Ku70 is closely related to lymphatic metastasis. Conclusion:1) It is suggested that PI3K, mTOR, Pokemon and Ku70 play an important role in NPC occurrence and development; 2) MTOR is related to pathological differentiation and prognosis; 3) Pokemon is closely related to pathological differentiation, malignant degree, neoplasm staging, distant metastasis, may become a prognostic biomarker; 4) Ku70 is closely related to lymphatic metastasis, and may become a prognostic biomarker.
