Objective To investigate the anti-tumor and anti-metastasis effect of Qingfei Mixture in vivo.Methods The mouse model of ICR bearing H22 ascites tumor was established to investigate the anti-tumor effect of Qingfei Mixture and the mouse model of C57BL/6 bearing Lewis lung carcinoma was set to investigate the anti-tumor effect, anti-pulmonary metastasis and synergetic anti-tumor effect of Qingfei Mixture. Further, expression of MMP-9 was detected by real-time chain reaction (RT-PCR) and expression of CD34 and VEGF proteins were observed by EnVision two-step immuno-histochemistry staining in tumor tissues from the mice bearing Lewis lung carcinoma.Results1. The low, moderate and high dose of Qingfei Mixture groups (12.70g/kg,25.40g/kg, 50.80g/kg) as well as CTX (30mg/kg) positive control group significantly inhibited tumor growth in the mice bearing H22 ascites tumor and Lewis lung carcinoma, the inhibitory rate were15.86%,36.12%,16.30%,53.74% and 21.98%,43.10%,27.58%,56.89%, respectively. And the moderate dose group displayed the most potent anti-tumor effect in the three groups of Qingfei Mixture.2. Qingfei Mixture group (25.40g/kg) combined with CTX (30mg/kg) exerted more potent anti-tumor effect compared with the separation groups of Qingfei Mixture and CTX in the mice bearing Lewis lung carcinoma.3. The low, moderate and high dose of Qingfei Mixture groups (12.70g/kg,25.40g/kg, 50.80g/kg) as well as CTX(30mg/kg) positive control group significantly inhibited pulmonary metastasis in the mice bearing Lewis lung carcinoma, the inhibitory rate were 16.70%,45.83%,19.44% and 80.56%, respectively. And the moderate dose group displayed the most potent anti-pulmonary metastasis effect in the three groups of Qingfei Mixture.4. Qingfei Mixture could down-regulate the expression of MMP-9 protein in carcinoma tissues from the mice bearing Lewis lung carcinoma, compared with saline control group, the positive expression rate of the protein in moderate and high dose groups was significantly different in statistics (P<0.01).5. Qingfei Mixture could make VEGF and MVD less expression in carcinoma tissues from the mice bearing Lewis lung carcinoma. Compared with saline control group, the effect of the moderate dose group on the two proteins showed significantly different in statistics (P<0.01). Conclusion Our researches indicate that Qingfei Mixture was an effective anti-tumor agent as well as having anti-metastasis effect and synergism with CTX in vivo, and the effect of the moderate dose group was superlative. It's molecular mechanism of anti-tumor could be concerned with down-regulating the MMP-9, CD34 and VEGF expression as well as depressing vascular growth in tumor tissues.
