Objective:To further explores the pathogenesis of prophylaxis and treatment for MIRI by traditional Chinese medicine through the obeservation of the effectiveness of KNHX-Ⅱon myocardial tissue MMP-9 of lab rat that got myocardial ischemia reperfusion injury, and finally provides the objective basis for researching and developing an Chinese medicine which can effectively prevent and cure MIRI.Methods:A model of myocardial ischemia-reperfusion injury was established by left coronary artery ligation on the Wistar rats. And 50 rats were divided randomly into 5 groups with 10 in each:sham operation group, model group, KNHX-Ⅱhigh dose group, KNHX-Ⅱlow dose group and TongXinLuo group. Lead electrocardiograph was recorded before and after reperfusion, and myocardial tissue was taken for HE staining and determination of MMP-9 dosage by immunohistochemical when model establishing finished.Results:1. Compared with the sham operation group, the myocardial MMP-9 dosage increased obviously (P<0.01) in model group. Compared with model group, the MMP-9 dosage obviously decreased (P<0.01) in the treating groups. No significant difference was showed between KNHX-Ⅱhigh dose group and the TongXinLuo group (P>0.05).The dosage of myocardial MMP-9 of high dose group was obviously lower than that of the low dose group (P<0.01).2. By microscope observing, pathological changes such as inflammatory cell infiltration, myocardial ischemia and myocardial cell edema, necrosis, nuclear condensation and nuclear dissolution appeared in different degree in each experimental group, which was less serious than the model group.3. Compared with the model group,the amplitude of T wave in KNHX-Ⅱhigh dose group and KNHX-Ⅱlow dose group decreased significantly. Conclusion:These results suggest that KNHX-Ⅱcould effectively ease the expression of MMP-9 and the amplitude of T wave in the lab rat that got myocardial ischemia reperfusion injury, and attenuate reperfusion injury.The pathogenesis could be reducing the inflammatory cell infiltration, protecting the myocardial ultrastructure and restraining the myocardial cell edema, necrosis, apoptosis and myocardial suppression.
