ObjectiveObserve the effects of jiont synovial fluid to the tendon-bone healing of reconstructing ligaments, define the relativity between jiont synovial fluid and tendon-bone healing, provide basic theory for choosing operation method of cross ligaments reconstruction.Method1.Experiment treatmentUsed 40 New Zealand white rabbits for experiment, divided them into 4 groups randomly, each had 10 rabbits. Used autogenous blood-supplying semitendinosus for transplant and did ligaments reconstruction operation on the upper shinbone. Operated double knee of rabbits, left knee was set to jiont synovial fluid group, and right knee was set to no jiont synovial fluid, then breed four groups rabbits in cage for 2,4,8,8 weeks respectively.2. Observation indexes2.1 Histological anatomy observationSentenced group 1,2 and 3 rabbits to death respectively after breeding, got specimen from marrow canal of upper shinbone, After fixed by 10% paraform-aldehyde, decalcificated by 10% EDTA,embedded and sliced, then HE stained and Masson histochemistry staining, observed the histomorphology changes of interface.2.2 Analyzed cell population of interfaceIPP cell counted the HE staining slice of group 1,2 and 3 then observed the density and histology condition of fibroblast.2.3 Biomechanical test Sentenced group 4 rabbits to death after 8 weeks of operation, measured the max tensile strength burden, breakpoint statistics and tensile strength bias curve of samples by 858 Mini BionixⅡbiomechanical test machine. 3 Statistics analysisUsed Single-factor analysis of variance and Paired t test statistics methods by SPSS 12.0 software.Result1. Compared the results of HE staining and Massonon staining slice at interface of marrow wayNo jiont synovial fluid group and jiont synovial fluid group showed great statistical significance of every time sections (P<0.05),and tendon-bone healing in no jiont synovial fluid group had better maturity than jiont synovial fluid group in the same time sect ion.2. Fibroblast density of HE staining sliceFibroblast density of HE staining slice in no jiont synovial fluid group was higher than joint synovial fluid group in every time sections,they showed great statistical significance(P<0.05),different time sections also had statistical significance (P<0.05).3. Biomechanical test resultsNo jiont synovial fluid group was better than jiont synovial fluid group at max tensile strength burden, breakpoint statistics and tensile strength bias curve in 8 weeks time section, they showed great statistical significance (P<0.05).Conclusion1. Ligaments reconstruction operation with no jiont synovial fluid circumstance can enhance the cambium cell grow into tendon-bone interface.2. Biomechanics function of tendon-bone healing with no jiont synovial fluid is better than jiont synovial fluid group.3. Tissue structure maturity of ligaments reconstruction operation with no jiont synovial fluid is better than jiont synovial fluid group.4. Jiont synovial fluid can interfere and restrain tendon-bone healing.5. When reconstitutiing ACL,we should avoid jiont synovial fluid permeate into bone marrow canal,this can improve tendon-bone healing and elevate operation effect...
