【Objective】 To investigate whether Hamstring tendon recombined with adenovirus vector carrying the human transforming growth factor-β1 genes could promote the tendon-bone interface healing of after rabbit’s anterior cruciate ligament reconstruction【Methods】Forty-eight New Zealand white rabbits were divided into three groups randomly. Using Hamstring tendon graft to Reconstuct the ACL.For the group A, using Hamstring tendon recombined with Ad vector carrying the TGF-β1 genes. For the group B, using Hamstring tendon recombined with Ad vector carrying nothing. For the group C, Hamstring tendon with Dulbecco’s DMEM. Four rabbits each group were killed at 2, 4, 8,12 weeks after operation,then evaluated by appearance studies and the patholody dyes(HE and Masson staining). we counted the number of fibroblasts and we obtained the Buark score in three groups.【Results】The observation of Anatomy. The rabbits were killed at 2 weeks, 4 weeks, 8 weeks and 12 weeks.Observed around the knee no signs of fever, swelling, wound infection. synovial significant proliferation,meniscus intact can be see in articular, reconstruction of theACL tension is good. At 2 weeks, between the transplanted tendon and the bone tunnel is still visible gap.At 4 weeks, the inner mouth of the bone tunnels have scar tissue. At 8 weeks, the external mouth of the bone tunnels have bone tissue. At 12 weeks, bone increase than 8 weeks, graft tendon and bone tunnel are closely connected. In histological evaluation. Group A:few fibroblasts and collagen fibers were found at the at 2 weeks after the surgery. At 4 weeks, chondrocyte-like tissue constituted tendon-bone interface,There were few collagen fibers and chondrocyte-like cells, the interface cleft could not be found easily. The fresh bone was appeared. At 8 weeks,there are chondrocyte-like shift-belt and chondrocyte-like cells ripe, Sharpey fiber appeared. At 12 weeks,dense connective tissues and calcified cartilage formed, regular Sharpey fibers were found, some areas was seen gradually blending from bone into the tendon grafts. Group B、C:at 2 weeks,few fibroblasts were found at the tendon-bone interface.At 4 weeks newly fibroblast and inflammatory cell could be found in it,the interface did not heal very much. At 8 weeks,There were a few fibroblasts, Sharpey fibers were not observed crossing this area.At 12 weeks, disorganised calcified cartilage and irregular Sharpey fiber were found, gradually blending from bone into the tendon grafts were not found. We counted the number of fibroblasts.We used One-way ANOVAand SNK test to analyze the data.The difference between group A and group B、group A and C were considered significant(P<0.05).The comparison between group B and group C were not obvious. Based on the fibrovascular tissues at the tendon-bone interface,we use Kruskal-Wallis H test to analyze the data of Buark score.Between group A and group B,group A and group C,the Buark scores were different(P<0.01). It means that the Buark score of group A was more than group B、C.【Conclusion】 Hamstring tendon recombined with adenovirus vector carrying the human transforming growth factor-beta1 genes could promote the healing of tendon- bone interface after anterior cruciate ligament reconstruction.
