[Objective] The aim of this study was to analyse the polymorphism and genetic background of D variants in sichuan, to fill the gap of the study of RHD variants ,as well as to provide a theoretical basis and some guidance of accurate identification of Rh blood group and clinical blood transfusion. the RHD zygosity was detected at the same time.[Methods] First of all, blood samples were identified by serological tests. D variants with weak D or partial D expression was screened using indirect anti-human globulin test (IAT) method. The adsorption and elution was used for testing of Del. Then, We studied the genotype of RHD by using polymerase chain reaction with sequence-specific primers(PCR-SSP),and all the samples with ambiguous results were sequenced. The number of RHD was also detected through PCR-SSP.[Results] Fifty-two samples were indentified as D variants by blood group serological tests. Following gene typing, RHD(G282D) was found 4 in weak D individuals, RHD(G277E) 1, RHD(L320L) 1, RHD(G263R) 1, DVI typeâ…˘was found 2 in partial D individuals. And also found 42 Del phenotype including one case of DELRHD (MlI)and 41 cases of DELRHD (K409K).A new alleles RHD(A237D)was discovered in our experiments, It has gotten the GenBank sequence number GU998825 .RHD heterozygosis test results showed that one cases of weak D15 and 9 cases of DELRHD (K409K) type specimens for the RHD+/RHD+homozygous, and others were RHD+/RHD-heterozygous type.[Conclusion] D variants population in Sichuan Province is rich in types and in different genetic backgrounds. [Objective]To investigate the RHD gene polymorphism among the RhD- individuals in population of sichuan.[Methods] The RhD negative samples were isolated from the blood donors by indirect antiglobulin test(IAT) or adsorption and elution method. We studied here the genotype of RHD by using polymerase chain reaction with sequence-specific primers(PCR-SSP),and one sample with ambiguous result was sequenced.[Results]The results showed that among 86 RhD- individuals,68 lacked the integral RHD gene,16 carried the RHD-CE(2-9)-D alleles,1 carried the RHD-CE(8-9)-D allele,and the last one carried the RHD (711DelC) allele.[Conclusion] RhD- individuals in Sichuan Province is rich in types and in different genetic backgrounds, the RHD Deletion is predominant in Sichuan area, and the RHD-CE(2-9)-D allele is also common besides it, some unusual genotypes liked RHD (711DelC) were also found in our experiments.
