In this experiment compared with extracting saponins from the stems, leaves and fruits of Momordica charantia L, with the Low-temperature condition. And crude saponin through AB-8 macroporous resin for purification, the purified samples by HPLC to check the components.This research mainly from two angles:anti-cancer and reducing blood glucose to describe the active substances of Momordica charantia L. Anti-cancer:Using MCF-7 human breast cancer cells as the model cells to study the effect of extractive ingredients from Momordica charatia L.on anti- MCF-7 by the MTT method. Hypoglycemic function: through cell experiments and animal experiments to study the active substances of Momordica charantia L's function. Cell experiments:from the target of restorative the (3 pancreas cells to research the repairing effects of extractive active substances from Momordica charatia L. on the HIT-T15 damaged by STZ; Animal experiments:Using the mice damaged by STZ as a model to study the effect of extractive substances from Momordica charatia L. to regulate blood glucose metabolism and stimulate insulin synthesis and secretion. Using glycogen assay kit to mensurate the muscle glycogen level, and mensurate fasting serum insulin levels by Iodine[125I] Insulin Radioimmunoassay Kit. The result:(1) Under the special extraction process and technical condition, we get the saponins from stem, leaves and flesh were 5.9%,13.6% and 2.01%. By AB-8 Macroporous resins system separating saponins, get a good results. At the optimum purification condition, Purified two times, saponins product contained more than 80%.Theck the components by HPLC:there are two components in the saponins of Momordica charantia L. saponin A close to Rgl from Ginseng and saponin A close to Rgl from Ginseng.(2) The saponins and polysaccharides substances extract from Momordica charatia L have certain value on restrain the rise of MCF-7. the result, the saponins extracted from the leaf of Momordica charatia L.had best effects, and saponins extracted from the pulp and stem of Momordica charatia L. followed, the worst effects was polysaccharide in Momordica charatia L.(3) The results show that active substances from Momordica charatia L. have significant role in repairing the pancreatic (3 cell line (HIT-T15) damaged by STZ. the effect relate with the concentration of drugs and drug ingredients. This experiment with seven concentration of extractive substances (50,100,200,300,400,500,600 ug/ml), only at the concentrations of 100ug/ml, each of the cell survival rate was highest; determined the concentration, saponins were the highest, followed by composite group, polysaccharides were the lowest.(4) Extractive substances from Momordica charatia L. with the high blood glucose mice as a model,play a role on lowering fasting blood glucose, increasing fasting plasma insulin secretion, alleviating tissue damage, lowering mortality and so on. lower blood glucose levels, saponins group and the polysaccharide and saponin compound group to be significantly higher than the polysaccharide group; Glycogen synthesis, the complex group were the best, saponin group second, and polysaccharide group worst; Increase insulin secretion, each group have significant effect, but the Saponin and composite group better than the polysaccharide group.
