Objective To observe the expression of Smad4, MMP-9 and CD34 in colorectal carcinoma tissue and discuss the correlation between the expression of Smad4, MMP-9, MVD and the clinical pathology parameter, and their role in the occurrence and development of colorectal carcinoma. And then, we analyzed the correlativity of the Smad4 and MMP-9 and their effect on the information of microvessel in colorectal carcinoma.Methods 47cases of the diverging colorectal carcinoma tissue, 23cases of normal tumor neighboring mucosa and 21 cases of adenoma were stained by Max Vision fast immunohistochemical method with antibodies against of Smad4, MMP-9 and CD34. And vascular endothelial cells were marked by CD34, then microvessel density was counted. The statistical software package SSPS 13.0 was used. Chi-square test or Fisher's Exact test, One-factor analysis of variance, Non-parametric rank sum test, T-test and Spearman correlation analysis were adopted, andα=0.05 was taken as test criterion.Results 1 The positive expression of Smad4 in colorectal carcinomas were much lower than it in normal tumor neighboring mucosa or adenomas, and there were statistical significance (P<0.05). The positive expression of MMP-9 in colorectal carcinomas were much higher than it in normal tumor neighboring mucosa or adenomas, and there were statistical significance (P<0.05). The MVD in colorectal carcinomas were much higher than it in normal tumor neighboring mucosa or adenomas, and there were statistical significance (P<0.01). And there was statistical significance (P<0.01) of MVD between normal tumor neigh- boring mucosa and adenomas.2 There were respective statistical significance between the positive expression of Smad4, MMP-9 and MVD of the Dukes C-D stage in colorectal carcinomas and these of the Dukes A-B stage (P<0.01). There were respective statistical significance between the positive expression of Smad4, MMP-9 and MVD of the group with lymph node metastasis and these of the group without lymph node metastasis (P<0.01). There were respective statistical significance between the positive expression of Smad4, MMP-9 and MVD of the group with distant metastasis and these of the group without distant metastasis (P<0.05). There were respective statistical significance between the positive expression of Smad4, MMP-9 and MVD of the non-serosal infiltration group and these of the serosal infiltration group (P<0.05). And there were respective statistical significance between the positive expression of Smad4, MMP-9 and MVD of the highly-moderately differentiated group and these of poorly differentiated group (P<0.05).3 In colorectal carcinomas, MVD increased along with the inactivation of Smad4, MVD mean of Smad4 positive group and negative group were 24.95, 31.20, and there was statistical significance between them(P<0.01). MVD mean of MMP-9 positive group was 30.52 and negative group was 25.25, there was statistical significance between them (P<0.01). In 47cases of colorectal carcinomas, 3 cases were negative expression of Smad4 and MMP-9, 5 cases were positive expression of both, 22 cases were negative expression of Smad4 but positive expression of MMP-9,and 17 cases were positive expression of Smad4 but negative expression of MMP-9. Statistical analysis revealed negative correlation of Smad4 and MMP-9 expression in 47cases of colorectal carcinomas (r=-0.659, P<0.01).Conclusions 1 The inactivation of tumor suppressor Smad4, the overexpression of MMP-9 and the formation of microvessel play fundamental roles in the occurrence and development of colorectal carcinoma.2 In colorectal carcinoma tissue, the expression of MMP-9 was negative correlation with Smad4, and MVD increased along with the inactivation of Smad4 and the overexpression of MMP-9, and so our results indicated that the inactivation of tumor suppressor Smad4 may facilitate the formation of microvessel through the effect on the expression of MMP-9, and then induce to the infiltration and diversion of the tumor.
