Objective This study was to explore diferential expression of miRNAs between Human nasopharyngeal carcinoma cells CNE1 and CNE1-LMP1 cells,and to predict potential targets of the diferentially expressed miRNAs. The first discusses LMP-1 for miRNAs expression spectrum influence of nasopharyngeal carcinoma cells as well as the impact of EBV oncogene LMP-1 in tumor promotion mechanism to do deeper research. For the diagnosis and treatment of nasopharyngeal carcinoma provide relevant basic data.Methods Human nasopharyngeal carcinoma cells CNE1 and transferred oncogene LMP-1 in nasopharyngeal carcinoma cell CNE1-LMP1 were cultured ,By immunohistochemistry and RT-PCR detection of LMP-1 expression in CNE1-LMP1,The total RNA was isolated and examined.The expression profiles of miRNAs in CNE1 and CNE1-LMP1 cells were detected with a miRNA microarray chip and compared Array.Potential mRNA targets wer epredicted by MiRanda,TargetScan, PicTar,and DIANA- microT software.Results miRNA has the specific expression level and the pattern in the different tumour cell, people can through to the tumor tissue miRNA expression and normal tissue expression of different comparative analysis, and comparative analysis of different tumor-specific identification of miRNA levels, thereby hel- ping to identify the early diagnosis and treatment of cancer. Because miRNA is adjustment member in the gene expression and the protein translation process, in tumor occurrence process regulative pivotal role, therefore, experts have predict- ted that the miRNA target molecule as a tumor biological therapy than the more effective coding genes as target molecules . LMP-1 in the EBV latent infection of nasopharyngeal have cancer genes characteristics ,plays animportant role in the nasopharyngeal carcinogenesis. At present in nasopharyngeal carcinoma EB -V encoded LMP-1 whether can affect the expression of miRNA, thereby regul- ates the expression of downstream target genes, causes changes in the biological behavior of tumor cells, the study has not been reported.According to amicrobar -rayy based screening.total 32 differential expressed miRNAs were found in 20 molecules expressing commonly, 8 were down-regulated and 12 were up regulat -ed in in CNE1-LMP1cells. The lower expression of difference is:hsa-miR-137, has-miR-151, hsa-miR-196a, hsa-miR-197, hsa-miR-202, hsa-miR-373,hsa-miR -488 and hsa - miR-213; In the 12 highly expressed miRNAs, there are six incre- ase over two or more: hsa-miR-19b, hsa-miR-17-3p, hsa-miR-22, hsa-miR-149, hsa-miR-150 and hsa-miR-188.131 potential target genes of mir- 19b were predi -cted by informatics analysis. For CNE1 - LMP1 cells were significantly differe- ntially expressed of miRNAs in bioinformatics analysis, screens the correspond- ding target genes. And found its target is very wide, involving oncogenes, cell di -fferentiation, transcription regulation, angiogenesis, signal transduction path wa -ys and so on.Conclusions The miRNA expression Profiles of CNE1 and CNE1-LMP1 cell are obtained.Some miRNAs may be involved in the pathogensis of breast cancer by regulating their target genes.
