Objective:To study the effect of XueBiJing(a concoction of Chinese herbs)on serum levels of high mobility group box protein 1 and heat shock protein 70 in severe acute pancreatitis rats.To investigate the protective effects of XueBiJing in severe acute pancreatitis.Methods:A total of sixty male SD rats were divided randomly into four groups:sham operation group(group A),SAP group(group B),low dose XBJ (4ml/kg) treatment group(group C), high dose XBJ (8ml/kg) treatment group(group D).SAP rat model was induced by 5% retrograde injection of Sodium taurocholate into the pancreatic duct, group C accepted intraperitoneal injection of XBJ at once after operation and repeated 12 hours later.Five rats in each group were sacrificed at 6,12,24h after operation.HMGB 1 and HSP70 serum levels were measured by ELISA.Results:1. There were no significant difference about the concentrations of HMGB1 at different time points in group A. There were no changes in serum concentrations of HMGB1 six hours after operation in group B, C and D, and comparing with group A, there was no difference. The concentration of HMGB1 gradually increased 12h after operation in group B, C, D, comparing with the same time point in group A, there were significant difference (P<0.05). The concentration at 12h and 24h in group C and D was significantly lower than the same time point in group B(P<0.05). The concentration at 12h and 24h in group D was significantly lower than the same time point in C group (P<0.05).2. The concentrations of HSP70 at different time points of group A were lower. The concentration of HSP70 gradually increased 6h after operation in group B, C, D, comparing with the same time point in group A(P<0.05). The serum concentration of HSP70 in group C and D was significantly lower than the same time poinst in group B(P<0.05). The serum concentration of HSP70 in group D was significantly lower than the same time points in C group (P<0.05).Conclusion:HMGB1 seems to act as a late mediator in severe acute pancreatitis. eHSP70 seems to act as a mediator in severe acute pancreatitis.Treatment with XBJ could obviously inhibit serum concentrations of HMGB1 and HSP70 in severe acute pancreatitis rats.XBJ possesses the protective effects in severe acute pancreatitis.
