ObjectiveInvestigate the therapeutic effects of the xinshenyihao on chronic psychological stress on serum-induced oxidative and damage in rat colon intervention. Explore the psychological stress of chronic rat model of colon injury in rats mechanism.Methods1. This experiment used 48 male SD rats.They were divided into 6 groups by random (8 per group).Divided into blank control group, model, western medicine group(WM) (paroxetine), on the xinshenyihao low, middle and high dose group.2. Chronic psychological stress model:at the beginning of every morning, the folder tail (1 hour), drilling barrels (2-3 hours), fixed limbs (2 hours), water (24 hours), cut fresh (24 hours) per day above a stimulating way, alternating 15 days. Treatment groups, the WM group will be prepared in accordance with 0.2mg/ml suspension paroxetine weight given lml/100g; medicine low, middle and high 2ml/100g weight given to each group (about 0.15g,0.30g,0.60g/100g crude drug). Control group given an equivalent model with distilled water gavage, continuing for 15 days.3. Compared with blank control group, model group, serum MDA content increased(P<0.05), compared with the model group, on the xinshenyihao serum MDA levels were lower (P<0.05), with WM group comparison, on xinshenyihao MDA content was the lower group (P<0.05).4.Blank control group, colonic mucosa layer, myenteric plexus of intestinal c-fos gene expression were negative, and the model have different levels of positive expression in the mucosal layer model group and the comparison between the control group(P<0.05),its expression is higher than the control group.Compared with model group, the treatment group the expression of c-fos-positive reduced(P<0.001). xinshenyihao low-medium-dose group the expression of c-fos-positive than the WM group(P<0.05).In the intestinal myenteric plexus model group compared with control group the difference was significant (P<0.05), its expression should be higher.Similarly, compared with the model group, the treatment group the expression of c-fos-positive reduction(P<0.001), and the dose on xinshenyihao group and WM there is no difference between groups (P>0.05).5.caspase-3 protein expression in the model group is higher than the control group (P<0.05),but expression of all the the xinshenyihao groups, WM group is below the model group (P<0.05), compared with the WM group, the expression of low-, Middle dose group is higher (P<0.05).Conclusions1. The psychological stress of chronic rat model is approximate to be in line with the symptom of Qiji disorder macro show. 2. Psychosomatic medicine paroxetine tablets and the xinshenyihao on the chronic psychological stress model of rat colon injury has a very good protective effect and xinshenyihao effects of high dose group is superior to WM group.3. xinshenyihao has reduced serum MDA content, SOD activity increased to improve the antioxidant capacity in rats, reducing lipid peroxidation injury in rats, its effect is superior to WM group.4. Chronic psychological stress model of rat colon c-fos, caspase-3 expression have a significant change, suggesting their involvement in stress-induced formation of colon injury, xinshenyihao, Paroxetine reduced the colon can be c-fos, caspase-3 expression.
