The Study On The Therapeutic Mechanisms Of The Prescription XINSHEN1HAO To Different Model Of Rat's Gastric Ulcer

Objective:Investigate the therapeutic effects of the XINSHEN 1 HAO on the different model of the Stomach Ulcer,and it's therapeutic mechanisms. This is good to excavate ancient prescription,and to provide a new idea and some experimental evidence in order to apply it in clinic better.Methods:1. This experiment used 100 male Wistar rats .They were divided into 10 groups by random(10 per group).There are two models.'â‘ Stress-induced Gastric Ulcer : the model group, the western medicine group(ranitidine group) , the small dosage group of XINSHEN 1 HAO.the middle dosage of XINSHEN 1 HAO, the large dosage of XINSHEN 1 HAO , and the contrast group .â‘¡gastric ulcer model by acetic acid damage: the model group, the western medicine group(ranitidine group), the middle dosage of XINSHEN 1 HAO, the fake operation group.2. Stress-induced Gastric Ulcer :We administered the 3mg/ml Ranitidine Capsules fluid to the Western medicine group ,lml/100g body weight (about 0.24g,0.48g,0.96g/100g crude drug) the XINSHEN 1 HAO fluid to the XINSHEN 1 HAO group. Buring ulcer by acetic acid: We administered the 3mg/ml Ranitidine Capsules fluid to the Western medicine group ,1ml/100g body weight (about 0.48g/100g crude drug) the XINSHEN 1 HAO fluid to the XINSHEN 1 HAO group, 1ml/100g body weight distilled water to the model group and the fake operation group for about 7 days.3. In the remedial process ,we observed the rat's appearance ,spirit ,behavior .7 days later ,we executed all the rats. Then we anatomized the rats , and separated the stomach.We select the pituitaries , to check their histopathological change. Using method of immunohistochemical assay to detect express of Bax,BCL-2,HSP70 of gastric mucosa.4. We analyzed the data by analysis of variance and q-test.Results:1. Stress-induced Gastric Ulcer:During the experiment,every group mouse outrage,bray,tug,worry and be easy to anger. Buring ulcer by acetic acid:After the operation, every group mouse become sag and normal after 3 days.2. It is succeed that adopting the method of water immersion and bondage to produce the mouse model of stress ulcer, stomach-tissue of different model group of morphology changed differently. Gastric wall of model group of degree of injury is most severity. To compare traditional Chinese medicine group with model group, has discrepancy statistical significance for UI(P<0.01). There are no difference between the traditional Chinese medicine group(P>0.05). It is succeed that adopting the method of buring ulcer by acetic acid to produce the mouse model of gastric ulcer, stomach-tissue of different model group of morphology changed differently. Buring ulcer by acetic acid: Bax positive cells in the model group were significantly more than those in Xinshenyihao-treat groups (P<0.01).4. Stress-induced Gastric Ulcer: Stress-induced Gastric Ulcer: Bcl-2 positive cells in Xinshenyihao-treat groups were significantly more than those in model group (P<0.01). Buring ulcer by acetic acid: Bcl-2 positive cells in Xinshenyihao-treat groups were significantly more than those in model group (P<0.01).5. Stress-induced Gastric Ulcer: Stress-induced Gastric Ulcer: HSP70 positive cells in Xinshenyihao-treat groups were significantly more than those in model groupConclusions:1. The traditional Chinese medicine compound Xinshenyihao have a good effection to different model of gastric ulcer.2. Stress-induced Gastric Ulcer:the expresstions of Bax,Bcl-2 and HSP70 in gastric mucosa have a distinctness change,it clews that they participate in the Gastric Ulcer. Xinshenyihao and ranitidine can adjust the expresstions of Bax,Bcl-2 and HSP70 to prevent the Gastric Ulcer.3. Buring ulcer by acetic acid: the expresstions of Bax,Bcl-2 in gastric mucosa have a distinctness change,it clews that they participate in the Gastric Ulcer. Xinshenyihao and ranitidine can adjust the expresstions of Bax,Bcl-2 to prevent the Gastric Ulcer.4. Xinshenyihao has the same effect mechanism to different model of gastric ulcer.
