Objective: Explore the relationship between serum ALT, pathological changes of liver, expression of P-selectin in serum of mice and different dose of venom, the poisoning time induced by Agkistrodon Halys Pallas venom.Methods: (1)Using two-factor factorial design, the 72 male Kunming mice of clean grade were divided into 9 groups(n=8) randomly. Snake venom was divided into three levels: 0-dose(0.0mg/kg), low-dose (0.5mg/kg), high-dose (1.0mg/kg); Poisoning time to set up three levels : 3 hours, 8 hours, 24 hours. (2)Collected serum and liver specimens, detected of ALT by biochemical analyzer, investigated pathological changes of liver, detected expression of P-selectin by Enzyme-linked immunosorbent assay.(3) Statistical Methods: experimental data using the SPSS 13.0 statistical software for statistical analysis, measurement data were presented as ( x±S), data analysis using factorial, correlation analysis using linear correlation analysis, P<0.05 regarded as statistically significant.Results: (1) The test group serum ALT value was significantly higher than the control group(P<0.05), high-dose group was higher than the low-dose group, the difference was significant (P<0.05).8h group is higher than 3h group(P<0.05),24h group is higher than 8h group(P<0.05).With the poisoning increased,the poisoning time to extend,the serum ALT is higher. (2) Through various periods of the statistical analysis of liver histopathologic scores, high-dose group is higher than the low dose group of serious diseases, the difference was statistically significant (P<0.05),liver histopathologic scores of 8h group than in the 3h group, and 24h group is higher than 8h group(P<0.05).With the poisoning dose increased,the time to extend,the liver histopathologic scores is higher. (3) Monitored mice serum expression of P-selectin was found that compared with the control group, experimental group of mice P-selectin expression was significantly increased, the difference was statistically significant (P<0.05).With the poisoning dose increased,the poisoning time to extend,expressing of p-selectin is higher. (4)correlation analysis: ALT value and liver histopathologic scores correlation analysis, showed a positive correlation (r=0.92,P<0.05);P-selectin expression and liver histopathologic scores correlation analysis, showed a positive correlation(r=0.98,P<0.05).Conclusions:(1)Agkistrodon snake venom can lead to acute liver injury of mice and elevated ALT, the more severe liver injury, the more ALT values increased significantly. (2)with acute liver injury of mice induced by Agkistrodon Halys Pallas venom,expression of P-selectin in serum of mice was increased,and the increase of the dose of venom and the poisoning time,the more expression of P-selectin ,showing a certain time,dose effect trend,suggesting that overexpression of P-selectin is one of the mechanisms of acute liver injury induced by Agkistrodon Halys Pallas venom.
