Objective:To build the mode of ALI,and to observe dynamicly the lung tissue morphology,the trend of changes of the PaO2,W/D,the content of protein in BALF, IL-1β,IL-10and the H2S/CSE system in agkistrodon halys pallas venom-induced acute lung injury in rats,and to investigate the mechanisms of ALIMethods:1.Fourty-eight male rats were randomly divided into two groups, control group and experimental group.The rats in control group were injected abdominally NS0.2ml,an the experimental group rats were injected Agkistrodon Halys Pallas Venom(1.50mg/kg).Every six rats in the two groups were respectively killed at1h、6h、12h and24h after administration of venom randomly.2.The PaO2,the W/D,the content of protein in BALF,the level of H2S in plasma,the activity of CSE in lung tissue and the contents of IL-1β and IL-10in plasma were respectively detected.The morphological changes of lung tissue were observed by light mircroscope.Results:1. The changes of morphological and the pathological scores of the lung injury:The structure of pulmonary alveoli were intact in control group.The structure of pulmonary alveoli were not altered at1hour after administration of venom compared with control group,and although the pathological scores of the lung injury was a little higher than the control group,there was no statistical mean(P>0.05).In experimental group6h,12h and24h,the lung tissues were injuried,the pulmonary alveoli were damaged,much exudate in the alveolar sacs and many inflammatory cells infiltrated.As long as the time last,the injuries of the lung were much more serious,and the pathological scores of lung injury were much more higher compared with the control group(P<0.05).2. PaO2and the correlation of PaO2and the pathological scores of the lung injury:In control group,the PaO2was not altered at1h、6h、12h and24h,(P>0.05). Compared with the control grouplh,the PaO2of the experimental group1h was decreased sightly,but there was no statistical mean(P>0.05);the PaO2of the experimental group was significantly decreased during6h、12h and24h after administration of venom compared with the control group(P<0.05or P<0.01).The analysis of correlation show that:the PaO2was impositively correlated with the pathological scores of the lung injury (r=-0.9444, P<0.01).3. The W/Dand the correlation of W/D and the pathological scores of the lung injury:In control group,the W/D was not altered at1h、6h、12h and24h,(P>0.05).The W/D was began to increase at1hour after administration of venom compared with the control group,but there was no statistical mean (P>0.05),and then was significantly increased during6hour to24hour after administration of venom compared with control group(P<0.01).The analysis of correlation show that:the W/D was positively correlated with the pathological scores of the lung injury (r=-0.9045,P<0.01).4. The content of protein in BALFand the correlation of the content of protein and the pathological scores of the lung injury:In control group, the content of protein in BALF was not altered at1h、6h、 12h and24h(P>0.05).The content of protein in BALF was began to increase at1hour after administration of venom compared with the control group,there was no statistical mean(P>0.05),and then was significantly increased during6h to24h after administration of venom compared with control group(P<0.01).The analysis of correlation show that:the content of protein in BALF was positively correlated with the pathological scores of the lung injury (r=-0.9085,P<0.01).5. The level of H2S in plasma and the activity of CSE in lung tissue and the correlation of the above two facts and the pathological scores of the lung injury:In control group,the level of H2S in plasma and the activity of CSE in lung tissue were not altered at1h、6h、12h and24h(P>0.05).The level of H2S in plasma and the activity of CSE in lung tissue were began to decrease at1h after administration of venom compared with control group(P<0.05),and then were significantly decreased during6h to24h after administration of venom compared with control group(P<0.01).The analysis of correlation show that:thethe level of H2S in plasma and the activity of CSE in lung tissue were both impositively correlated with the pathological scores of the lung injury (r=-0.9163, r=-0.943,P<0.01).6. the contents of IL-1β and IL-10and the correlation of the level of H2S in plasma and the activity of CSE in lung tissue:In control group,the contents of IL-1β and IL-10were not altered at1h、6h、12h and24h(P>0.05). the contents of IL-1β and IL-10were significantly increased at1hour after administration of venom compared with control group(P<0.01).During6hour to24hour after administration of venom,the contents of IL-1β and IL-10were decreased compared with venom1h group while were increased compared with control group(P<0.05).The analysis of correlation show that:thethe level of H2S in plasma and the activity of CSE in lung tissue were both impositively correlated with the contents of IL-1β and IL-10(r=-0.8837、r=-0.8486、r=-0.9409、r=-0.9410,P<0.01).Conclusions:1The agkistrodon halys pallas venomIn can induce ALI in rats.2The decrease of the H2S/CSE system might be played a role in the physiopathologic process of ALI induced by venom.3In the progress of ALI in rats induced by agkistrodon halys pallas venom,the inflammatory cell factors may be released abundantly through some way striked by the decrease of the H2S/CSE system,which made the injuries of the lung tissue much more serious.
