Objective:①To investigate the expression of cyclooxygenase-2(COX-2) and inducible nitric oxide synthase(iNOS) in primary pterygia through immunohistochemistry, and to understand the correlation between the expression of COX-2 with iNOS in primary pterygia;②To study the possible effect apoptosis plays in the incidence and development of pterygium.Methods:56 samples of primary pterygia were collected for the present study. These samples were cut off from surgical operations in ophthalmology section of First Affiliated Hospital of Medical College of Shihezi University from 2008 to 2009.20 samples of normal conjunctiva were selected. All samples were diagnosed. The pathological specimen was cut in the thickness of 3um. Immunohistochemical Elivision staining for COX-2,iNOS were made. The results of immunohistochemistry were determined by the intensity of staining and the number of positive cells. The expressions of COX-2,iNOS in primary pterygia and normal conjunctiva were analyzed. And the expression of apoptosis in primary pterygia were determined by terminal deoxynucleotidyl transferase mediated 2'-deoxyuridine 5'-triphosphate-biotin nick end labeling(TUNEL) method respectively.Results:1 The expression of COX-2 in primary pterygia was up-regulated which had significantly different (P<0.05) from that of both resting stage and active stage and normal conjunctiva. The expression of iNOS in primary pterygia was up-regulated, which was significantly different (P<0.05) from that of both resting stage and active stage and normal conjunctiva.2 There are expression of apoptosis both in resting stage and active stage in primary pterygia.Conclusion:1 Either COX-2 or iNOS may accelerate the genesis or progress of primary pterygia.2 There may be a cooperation between COX-2 and iNOS in the genesis and progress of primary pterygia.3 Apoptosis caused by COX-2 and iNOS relate to the pathogenesis of primary pterygia.
