To detect the expression of HER-2 gene more exactly, this study tried to examine the amplification of HER-2 by combining TMA (tissue microarray) with FISH technology in breast cancer.Methods:1. 30 paraffin-embedded tissues of invasive ductal carcinoma of the breast were collected.2. FISH and IHC staining were performed to detect HER-2 gene amplification and protein expression in these cases. And then the relevance between the results of the two techniques was compared.3. The tissue chip was prepared by collecting all tissues from 30 cases, and the FISH test was further performed in the tissue chip to detect the amplification of HER-2.Results:1. In 30 cases, FISH test showed that HER-2 was detected by FISH in 8 cases, and was not detectable in 22 cases; IHC showed that HER-2 expression was: (3+) 3 cases, (2+) 20 cases, (1+ and 0) 7cases. FISH identified gene amplification in 66.7%of IHC score 3+ tumors(2/3) and in 30%of IHC score 2+ tumors(6/20),thus the concordant ratio between IHC and FISH was 73.3%(22/30).The two results showed significant correlation(P<0.01).2. In 13 cases, HER-2 amplification was detected in 5 cases by tissue chip and FISH test; however, HER-2 amplification was not detectable in 8 cases. Its concordant ratio with the usual sections' FISH results was 76.9%(10/13) and the two results showed correlation(P<0.05).The other seventeen cases had no satisfactory results for tissue sloughing or discontented disposal.3. TMA and FISH technology were combined together to detect the amplification of HER-2 gene in breast cancer. It was suggested that this method could completely preclude the FISH-positive cells from the carcinoma in situ of the breast to specifically definite the amplification of HER-2 gene in the invasive carcinoma of breast cancer.Conclusion:The FISH and IHC testing results of HER-2 status of breast cancer could have relevance. The FISH test results of the tissue microarray and the general sections may be relevant.
