Objective:Explore the serum concentration of XCLl.y interferon.T-cell subsets and HBV-DNA load in HBV-infected patients and its clinical significance.to provide reference that XCL1 participate the hepatitis B chronic pass.Methods:The serum concentration of XCL1 was detected by enzyme linked immunosorbent assay (ELISA);Peripheral blood T-cell subsets were detected by flow cytometry(FCM);liver function was assayed by automatic biochemistry analyzer;hepatitis B antigen/antibody semi-quantitative index was detected by Time-resolved fluorescence analyzer;HBV DNA was detected by Real-time fluorescent quantitative PCR.Results:1.Serum concentrations of XCL1 in acute hepatitis B group (n=20),mild chronic hepatitis B (CHB) group (n=30),moderate CHB group (n=30),severe CHB group(n=30) and normal control group(n=30) were (17.374±3.059)ng/ml,(11.128±1.870)ng/ml,(12.409±2.244)ng/ml,(14.650±2.864)ng/ml,(8.289±1.820)ng/ ml respectively;IFN-y concentrations were(45.011±4.504)pg/ml,(36.340±5.558)pg/ml,(33.724±6.341)pg/ ml,(35.034±7.718)pg/ml,(19.975±2.916)pg/ml.As the severity of liver inflammation,XCL1 concentration increasing. In acute and chronic hepatitis B group serum concentration of IFN-y were significantly higher than the normal control group (P<0.05),and acute hepatitis B group has the highest concentration of IFN-y. 2.The serum level of CD4+T cell and CD4+T/CD8+T in acute and chronic hepatitis B group compared with the normal control group were significantly different (P<0.05).3.XCL1 was positively correlated with ALT and DBIL, related coefficients were 0.464(P<0.01) and 0.462(P<0.05). IFN-y was positively correlated with ALT and DBIL, related coefficients were 0.326 (P<0.01) and 0.191(P<0.05).The serum live of XCL1,IFN-y were positively related(r=0.714 P<0.01).4. XCL1 were negative correlated with CD4+T cell and CD4+T/CD8+T, related coefficients were-0.251 (P<0.01) and-0.432 (P<0.01), XCL1 was positive correlated with CD8+T cell, related coefficients was 0.296 (P<0.01).Serum IFN-y concentration was positive correlated with CD8+T cell, related coefficients was 0.238(P<0.05), and IFN-y were negative correlated with CD4+T cell and CD4+T/CD8+T, the correlation coefficient r were-0.334(P<0.01) and-0.338 (P<0.01).5. Serum concentrations of XCL1 in low viral load group,high-load group and normal control group were (15.172±3.146) ng/ml,(12.974±3.211) ng/ml,(8.289±1.820) ng/ml respectively, IFN-y concentration were(38.771±6.429)pg/ml,(36.125±7.598)pg/ml,(19.975±2.916)pg/ml respectively.The level of XCL1 and IFN-y in low viral load group and high-load group were higher than normal control group (P<0.05).Conclusions:1.The concentration of XCL1 was positive correlated with ALT and DBIL concentration, XCL1 may involved in the hepatitis B virus infection after liver inflammation injury.2. The serum live of XCL1 and IFN-y were positively related, by changing the concentration of IFN-y XCL1 influence the body's clearance of HBV.3. IFN-y mainly involved in the body's immune clearance of hepatitis B virus. XCL1 and IFN-y played a synergistic role in the immune regulation of hepatitis B virus-induced injury and involved in the process of chronic hepatitis.
