| Objective: Human leukocyte antigen (HLA) class II alleles have been associated with an increased or decreased risk of developing cervical cancer through infection with oncogenic forms of human papillomavirus (HPV). To verify whether HLA-DRB1*1501,-DQB1*0602 is related to HPV infection and invasive squamous cell carcinoma of cervix (ISCC) in Xinjiang Uigur and Han women.Methods: 287 cervical cancers (Uigur: 192; Han: 95) and 312 healthy controls (Uigur: 218; Han: 94) were analyzed for HLA typing by using Polymerase chain reaction-sequence specific primer (PCR-SSP) method. All samples were detected 16 and 18 subtypes of HPV, by using Polymrase chain reaction (PCR).Results: Our results showed that DRB1*1501 allele (OR,0.305;95%CI,0.115-0.813;p=0.013)and DRB1*1501-DQB1*0602 haplotype (OR,0.274;95%CI,0.086-0.874;p=0.021) frequency were less than healthy control in Han women. A significant relationship was found between DQB1*0602 (OR,0.482;95%CI,0.325-0.716;p=0.000) and decreased risk for cervical cancer in Uigur women. Similar tendencies were observed for DQB1*0602 with HPV16 positive cervical cancers (OR,0.550;95%CI,0.362-0.837;p=0.006). And we first report that DRB1*1501 may confer susceptibility to HPV16 infection among Uigur women. We also find that the positive rate of HPV 16/18 in cervical cancer were significantly higher than that in control group both in Uigur and Han. And the positive rate of HPV16 in Uigur were much higher than that in Han women(χ~2=6.861, p=0.010).Conclusion: These result indicate that DRB1*1501 and DRB1*1501-DQB1*0602 haplotype may contribute to the resistance to the development of cervical cancer in Han women, and the same contribution was made by DQB1*0602 for cervical cancer in Uigur. And DRB1*1501 may increase the susceptibility of HPV infection of Uigur women, along with the higher positive rate of HPV infection may explain phenomenon of high incidence of cervical cancer in Uigur women. The study suggests that specific HLA class II alleles and haplotypes may influence the immune response to HPV infection and affect the risk of cervical cancer in Xinjiang Uigur and Han women. |
