Purpose To evaluate the role of REV3 gene in tumorigenesis and development by down-regulating the influence of REV3 gene in the expression of colon carcinoma cell genetic information and the reproduction of transplantation tumor.Methods We experimentally suppressed the REV3 expression in human colon cancer cells (SW480 and COLO-205) by the interference RNA technology (RNAi) as monitored by real-time RT-PCR, cells that had low express rate were selected as case group. We detected the indexes of cell growth curve, MTT, micronuclei and sister chromatid exchange in order to analysis the influence of REV3 in genetic informational expression such as tumor cell growth and genetic instability. Observed and analyzed the difference of reproduction of transplantation tumor model through transplanting colon carcinoma cells with down-regulated REV3 gene into nude mice.Result Compared with untreated or mock-treated cells, ablation of REV3 significantly reduced cell growth rate, micronucleus formation and the frequency of sister chromatid exchange (P<0.05), whereas differences between untreated and mock-treated controls were insignificant. Indicating that the low expression of REV3 might affect the growth and proliferation of colon cancer cells (SW480 and COLO-205), and contributed to the interference of tumor proliferation formed in nude mice.Conclusion REV3 gene contributes to tumorigenesis and tumor development directly, and we can interference the development of tumor by regulating its expression appropriately.
