Objective: To study the effect of wasabi extract on the proliferation of colon cancer cells SW480 and the expression of tumor suppressor genes P16INK4a ,and get the preliminary understanding of the mechanism.Methods: After the 48h stimulation of different concentrations of wasabi extract, the inhibition effect of human colon carcinoma cells SW480 was measured by MTT assay, and the IC50 of Wasabi extract was also got, and the morphology changes of SW480 cells was observed under the phase contrast microscope, and cell apoptosis and cycle distribution changes were observed by flow cytometric, and mRNA expression changes of tumor suppressor gene P16INK4a was detected by RT-PCR.Results: Proliferation of human colon carcinoma cells SW480 was inhibited by Wasabi extract and had a dose response relationship with Wasabi extract, and cell apoptotic bodys were obvious observed under the phase contrast microscope, and the cell prophase apoptosis had a positive correlation with Wasabi extract concentration and most of the SW480 cells were blocked at G2 phase by Wasabi extract under flow cytometric , and the results of RT-PCR show that mRNA expression of tumor suppressor gene P16INK4a was reduced after 48h treatment of Wasabi extract.Conclusion: Wasabi extract could inhibit proliferation and induce apoptosis of human colon carcinoma cells SW480, by inhibiting expression of tumor suppressor genes P16INK4a which lead to cell cycle through G1 phase and arrest at G2 phase.
