Diabetes is a frequently encountered disease for old folks, dynamic, continuous and accurate detection of human blood glucose concentration is the key for prevention and treatment of the diabetes. At present, the detection methods of human blood glucose concentration mainly include invasive, minimally-invasive and non-invasive. Among the three methods, invasive detection bases on phlebotomized from fingertip, this technology made the patients feel pain, and also can't achieve dynamic and continuous detection. The method of non-invasive blood glucose monitoring technology in the world is still in the research stage, there is a large distance away from the clinical application. Therefore, minimally-invasive blood glucose testing technology becomes research hotspot. The minimally-invasive human glucose detection technology was studied by our group. Though interstitial fluid transdermal extraction, it can estimate the blood glucose concentration through detecting glucose concentration in the interstitial fluid by SPR.The technology of detecting glucose concentration was studied in this paper. And through introduction GGBP which can absorb the glucose specially, the experiments of immobilizing GGBP and measuring glucose concentration were carried out, various factors in the experiment were analyzed, a portable glucose detection instrument was developed. The work accomplished in this paper mainly includes the following:(1) Current development situations of blood glucose detection were studied, the principle, development situations and application of SPR were expounded, and features, preparation methods and fixation methods of GGBP were discussed.(2) The protein binding of the amine coupling method was studied, and the experiments of immobilizing GGBP by amine coupling and measuring glucose were carried out. The experimental results show that: the measuring results of based on amine coupling are better than the results of direct measurement. The glucose detection resolution could reach 0.5mg/dL, and it has good linearity relationship when the glucose concentration is around 0.5mg/dL to 5mg/dL. (3) The protein binding of the thiol coupling method was studied, and the experiments of immobilizing GGBP by thiol coupling and measuring glucose were carried out. The experimental results show that: the measuring results of based on thiol coupling are better than the measuring results of based on thiol coupling. It has good linearity relationship when the glucose concentration is around 0.1mg/dL to 10mg/dL. Though a lot of experiments, the stability and repeatability of the measurement were verified.(4) Various factors in the experiment including pH value, protein concentration, protein activity, protein preparation process and temperature were analyzed.(5) A portable minimally-invasive human glucose detection instrument was developed, and verification experiments of the instrument's feasibility were carried out in this paper, the existing problems of this instrument were analyzed and the improvement advices were proposed.
