Objective: to search of PI3K pathway inhibitor LY294002 on paclitaxel chemotherapy resistant ovarian cancer cells and the connection with the spindle checkpoint protein Mad2. Measures: Use of Western-Blot to detection of ovarian cancer cell line A2780 and A2780T after LY294002 and paclitaxel application Mad2 protein expression. Use Realtime-PCR to detection of ovarian cancer cell line A2780 and A2780T after paclitaxel and LY294002 application PI3K, PI3K, Mad2 of mRNA, and the application of flow cytometry in ovarian cancer cell line A2780 and A2780T after paclitaxel to detection cell cycle and apoptosis.Results: PI3K inhibitor LY294002 significantly enhanced ovarian cancer cell line A2780 and A2780T sensitivity to paclitaxel, apoptosis was significantly higher than the control. LY294002 significantly decreased the role of ovarian cancer the expression of PI3K and Mad2.Conclusion: PI3K inhibitor LY294002 inhibit PI3K, so that the sensitivity to paclitaxel in ovarian cancer cells significantly increased, played a role in reversing resistance to chemotherapy. At the same time, significantly reduced its spindle checkpoint protein Mad2 expression, consistent with PI3K obvious, and with LY294002 dose and time related. Therefore, LY294002 reversed the mechanism of resistance to chemotherapy may be played by Mad2 some effect. Objective: To investigate the expressions of Mitotic Checkpoint Protein Mad2 in ovarian cancer,explore the mechanism of initiation and prognosis of tumors。Methods: Immunohistochemical staining was used to investigate the eXpressions of Mad2 of 57 ovarian cancer cases. The correlation between the differences of the protein expressions and the biological features was analyzed.Results: Mad2 expression in ovarian cancer was significantly higher than benign ovarian tumors and normal ovaries (74%, 67%, 50%), the difference was statistically significant (P <0.05). And the positive rate of high and low with the grading and staging is enhanced. Objective To Study The perioperative management patients of gynecological patients with Lacunar cerebral infarction .Method A total of 11 cases of gynecological patients with lacunar cerebral infarction were diagosed and treated from 1991 to 2009 in Tongji Hospital of Tongji Medical College of HUST. Clinical and pathological data were reviewed and analyed.Result 11 cases of gynecological patients with lacunar cerebral infarction through perioperative period safely ,no postoperative complications, satisfaction with treatment.Conclusion gynecological patients with lacunar cerebral infarction, for their disease characteristics to do a good job before, during and after the treatment can be effective in reducing the incidence of complications.
