Herba Capsella (Ji Cai) is from Capsella bursa-pastoris (L.) Medic. (Cruciferae), which has the functions of cleaning away heat evil and promoting diuresis, calming the liver and improving acuity of vision, cooling blood to stop bleeding. Capsella bursa-pastoris, in which flavonoids are the mian active constituents, has action to treat edema of nephritis, odynuria, hemafecia, menorrhagia, conjunctival congestion, diarrhea, dysentery, chyluria and hypertension and so on. To provide the basis for quality assessment and application of Capsella bursa-pastoris, research on chemical reference substances, quality control method and the preparation technology of its extract studies were carried out for the first time.The review on chemical constituents, pharmacological activities, clinical application and the research progress of Capsella bursa-pastoris were presented.Research on chemical reference substances of Herba Capsella Two compounds were separated from Capsella bursa-pastoris (L.) Medic, with solvent extraction and column chromatography such as Macroporous adsorption resin, polyamide, Sephadex LH-20 chromatography and semipreparation HPLC, and were identified as isoorientin (1) and isoorientin-2″-O-α-L- arabinopyranosyl (2), both of which were isolated from this genus for the first time. Analyzed by HPLC, their purities were 98.6% and 99.3% respectively, which met the requirement of the standard substances.Research on quality control method of Herba Capsella Systematic researches and assessment on Herba Capsella collected from 21 different areas, including identification, limit test and assay, were performed. It provides basis for scientific and reasonable quality control of Capsella bursa-pastoris. Microscopic identification was carried out. The method of TLC identification of Capsella bursa-pastoris was set up, using isoorientin-2″-O-α-L-arabino-pyranosyl as refernce substance. HPLC fingerprint was established, in which two peaks were identified respectively as isoorientin-2″-O-α-L-arabinopyranosyl and isoorientin.Water, total ashes, acid insoluble ash and ethanol-soluble extractives were determined.A reliable and accurate high-performance liquid chromatography (HPLC) method was developed for the quantitation of isoorientin-2″-O-α-L-arabinopyranosyl in the extract. Analytical curve for isoorientin-2″-O-α-L-arabinopyranosyl was in the range of 21.0~210.0μg/mL with r=0.9998, recovery is 99.77%. The method was simple, accurate, rapid and specific, which can be used for the quality control of the extract.Research on preparation technique of Herba Capsella The extraction technic of Capsella bursa-pastoris was confirmed by L9 (34) orthogonal design with total flavonoids and isoorientin-2″-O-α-L-arabinopyranosyl as criterions. The conditions of the extraction technic were A2 % ethanol, immersed overnight, C2 multiple of A2 % ethanol refluxing extract for D2 times, B2 mins every time. III type macroporous resin was used in the separation and purification from Capsella bursa-pastoris by the comparison of the adsorption and desorption capacity of three kinds of macroporous adsorption resins with the content of the total flavonoids as index. The optimum technic was that the sample solution was loaded on the column (d/H = 1 :H2) with the adsorption rate S2 BV/h, and then eluted with F2 % ethanol with the desorption rate W2 BV/h. Under this condition, a desorption solution of approximate M BV completely desorbed total flavonoids from III resin. The content of total flavonoids is 62.9%.
