Objectives: To investigate the effects of multiwall carbon nanotube (MWCNT) andmultiwall carbon nano-onion (MWCNO) on the protein expression profiles of human skin fibroblasts (HSF) using proteomic techniques.Methods: Protein expression profiles were analyzed using 2 dimensional gelelectrophoresis, and differentially expressed proteins were identified by Matrix-assisted lasor desorption ionization-Time of Flight mass spectrometry (MALDI - TOF - MS). The expression ofβ-actin was evaluated by immunofluorescent microscopy.Results: After low concentrations (0.06 mg·L-1 MWCNT and 0.6 mg·L-1 MWCNO)and high concentrations (0.6 mg·L-1 MWCNT and 6 mg·L-1 MWCNO) treatment for 24 h, the expression levels of 22 proteins were changed. Among them, 10 proteins, includingβ-actin, were identified. Immunofluorescent microscopy also revealed the expression changes ofβ-actin.Conclusion: MWCNT and MWCNO can affect cellular protein expression. Proteomic technique can be applied in the study of nanotoxicology, which would provide useful information for the further study of molecular mechanisms. |
