Objective: To investigate the clonality composition of the entire lesion, Polygonal and surface cuboidal cells of so-called pulmonary sclerosing hemangioma (PSH) pulmonary sclerosing hemangioma (PSH), and their relationships. Methods: The PSH entire lesion, polygonal and surface cuboidal cells were microdissected from 29 female patients with PSH, respectively. Genomic DNA was extracted, pretreated by methylation-sensitive restrictive endonuclease Hha I and Hpa II, and then X– chromosome - chained androgen receptor (AR) gene and phosphoglycerate kinase (PGK) was amplified via nested polymerase chain reaction (PCR). Length polymorphisms and clonality of AR gene product were shown by denatured polyacrylamide gel electrophoresis and silver staining, and that of PGK gene product were shown by 2 % gel electrophoresis.Results: Microscopically, typical histopathological features were observed in all the 29 cases. The tumor cells arranged in solid and papillary, and hemangioma could be found in some cases. The cells in solid region were round or polygonal, and their cytoplasms were acidophilia or slightly transparent. Mitotic figures was rare. The surface of papillary region were covered with simple cuboidal epithelium. Cells in the solid region were in similar size. Clonality assay showed AR gene polymorphism in 23 cases,PGK gene polymorphism in 7 cases.Following digestion by HhaI,HpaⅡ, density of one band in all samples which had polymorphism significantly decreased or disappeared, but remained in the other band, which confirmed they were neoplastic proliferation.Conclusion: Polygonal cells and surface cuboidal cells of PSH lesion have the same monoclonal proliferative pattern. This suggests they were the neoplastic parenchymal cells.
