Jaw fractures except the performance of hemorrhage, bone fracture shift, paraesthesia and function barrier, the ache and swelling also are the extreme matter of concern by clinician. Therefore, in treating patient's Jaw fracture, the Steroidal anti-inflammatory drugs (dexamethasone) are often used for inhibiting the inflammatory response, alleviating the local swelling. And Non-steroidal anti-inflammatory drugs (indometacin) are often used for alleviating the pain. However, while the hormone plays the anti-inflammation role, it has what impacting on the bone fracture healing not yet to expound until now, and the painkillers on impacting fracture healing is still controversial. This experiment establishes the rat mandibular defect healing model of self-limited basis, using the average gray-scale X-ray measurement, morphology, total collagen staining method, evaluating the effect of steroidal anti-inflammatory drugs dexamethasone and non- steroidal anti-inflammatory drugs indometacin on impacting mandibular bone defect healing.Methods: SD rats were taken the operation for incomplete fracture of hibateral mandible penetrating from buccal side to lingual side. The rats were randomly divided into two groups─the experiment group was administered dexamethasone sodium phosphate, and the control group was administered physiologic saline; the experiment group was administered indometacin, and the control group was administered physiologic saline. All rats were administered for five days starting from the day of surgery. The animals were killed on the first, third, sixth, tenth and seventeenth day after five days'administration, and taken out of hibateral mandible. After X-ray examination, PLP fixative fixed, 10% EDTA decalcified, paraffin-embedded sections, conventional HE staining and total collagen staining, the effect of dexamethasone and indometacin was assessed by analyzing the proportion of total collagen.Results: The average gray-scale analysis showed that defect healing in the control group were higher than that in dexamethasone experimental group. Experimental and control groups between the significant difference (P = 0.016), with the difference in healing time and more obvious; Histological observations revealed that the healing in the experimental group was significantly slower than that in the control group. The proportion of total collagen showed that the defect healing in the control group was higher than that in the experimental group. The experimental group and the control group had the significance difference(dexamethasone group:P=0.013; indometacin group:P=0.018). The proportion of total collagen analysis had non-significance difference between dexamethasone experimental group and indometacin experimental group(P=0.223).Conclusion: 1) The mandibular bone defect by the size of 1mm×3mm in rat can heal voluntarily. 2) In inflammatory stage, applying therapeutic dose of Steroidal anti-inflammatory drugs dexamethasone and non- steroidal anti-inflammatory drugs indometacin can delay the healing of mandibular bone defect.
