Objective:Essential hypertension is a major public health concern worldwide where its prevalence accounts for various cerebrovascular diseases. There are many factors that can cause essential hypertension, and the inherent factor and the environment factor are the two main factors which they interact each other and lead to essential hypertension. Some studies of molecular epidemiology in recent years have been devoted to exploring the association and potential mechanism between hypertension and angiotensiongen (AGT) gene-variation. However, the results of published reports derived from different, mostly small, subpopulations and regions were not consistent with each other. Here, a case-control study was used to investgate the association of M235T of AGT with the hypertension in Shanxi Province.Methods:146 patients with essential hypertension and 182 normotensive subjects were involved in this study. The criterion is 2005 Chinese Hypertension Guidelines and subjects have been diagnosed as hypertensive person. We registered the clinical data and detected biochemical indicator of each subject.2ml anticoagulated blood was obtained for genome DNA extraction, then waiting for the collecting to measure at the same time. Genomic DNA was isolated from white blood cell. Mutagenically separated polymerase chain reactions (MS-PCR) was used to detect the variant in the exon 2 of AGT gene. SPSS 13.0 software pack was used to analyze all the data. All the continuous variables were expressed as mean±SEM. Gene counting method was used for gene frequency, and Chi-square test was used for analyzing allele frequency and analysis of categorical data. Students-t test was used for group comparison. A P-value of less than 0.05 was considered to be statistically significant.Results:1.There are no significant differences in the common clinical data and biochemical indicators between the two groups (P>0.05), including sex, triglyceride, total cholesterol, high dense lipoprotein cholesterol, low density lipoprotein cholesterol.There are a differences in the age (P<0.05).2. Hardy-Weinberg equilibrium for the genotype distribution was assessed as before, the crowd which was selected was representative (P>0.05).3. There was no association in the three genotypes and allele frequencies between hypertensive and normotensive subjects(P>0.05).4. The odds ratio for those exposed to TT genotype of AGT gene was the highest. (MM: OR=0.415, CI:0.205-0.841, P=0.015; T: OR=0.306, CI:0.151-0.617, P=0.001)。Conclusion:The AGT M235T polymorphism might be associated with essential hypertension in Shanxi Province. Patients with 235TT genotype have higher chance to get essential hypertension.
