Objective: By observing the general condition, tumor size and the expression of VEGF and MVD after the endostatin combined with radiation therapy on mice, we researched synergistic effect of Endostar combined radiotherapy on renal cell of xenografts carcinoma in mice.Methods: We successfully established×enograft model of female BALB /C mice with Renca renal carcinoma. It is about 15 days after inoculation. Until the volume of tumor reach about 0.5cm~3, we randomly divided the 48 female BALB/ c mice with Renca renal cell carcinoma into 4 groups (n = 12): (1) control group; (2) alone Endostar (10mg/kg/d) group; (3) radiotherapy alone group (4) radiotherapy + Endostar (10mg/kg/d) group. Endostar was used for 14 days at the beginning of radiation. Radiotherapy dose was 5Gy / times. Every other day mice were radioed for a total of 4 times. Every day we observed tumor condition .We measured maximum diameter of the tumor (A) and the maximum width by perpendicular (B) every other day and drawn tumor volume growth curves in the end.After the mice were killed when the treatment was over, we measured the size, Immunohistochemistry assay was used to determine the expression of MVD and VEGF of each group.Results: (1) 48 female BALB /C mice of tumor model were successfully established. (2) Tumor volume of each group increased, but the group of combination was the slowest. Statistical analysis of tumor volume showed significant differences (P <0.05). (3) HE staining showed that the combination group showed a large tumor necrotic area compared with other groups. (4) Statistical analysis of the expression of VEGF and MVD showed: Combined group was significantly less than other groups (P <0.05).Conclusion: The recombinant human endostatin combined with radiotherapy has a synergistic effects on mice with Renca renal cell carcinoma and can significantly improve the value of treatment of mice with Renca renal cell carcinoma.
