Objective:To observe the effects of AMD-zhikangyin on the rat model of nonalcoholic fatty lier disease (NAFLD)with high-fat diet;To investigate the influence of liver X receptor alpha (LXR alpha) and their downstream gene sterols regulating elements combined with protein lc (SREBP-1c) in fatty liver tissue;To discusses the therapeutic effect of superfine zhikangyin on NAFLD and its possible mechanism and provide a reference for clinical drug treatment.Methods:68 SD rats(male and female) randomly divided into two groups: 12 in the blank group which were fed with ordinary diet,and 56 in the high-fat diet group,which were fed on improved high—fat diet,after raising six weeks,8 rats were randomly drew off from each group(from the normal group only 2 rat and only 6 model rats)to take liver do pathological tip with celiac anesthesia to identify the NAFLD model,the remaining 50 high-fat diet rats by randomly divided into 5 groups:the modell group,the ultraicro zhikangyin in low dose group(i.e.drink low dose group),the ultramicro zhikangyin in middle dose group(i.e.middle dose group),the ultramicro zhikangyin in the high-dosage groups (namely high dose group),kezhi capsule group (positive medicine group), each group are 10 rats,Animal in the blank and model group were poured into stomack Nomal Saline,the other groups were poured into respectively drug at theirselves dose.After six weeks later, put to death, taking on the eve of the forbidden water fast blood and liver tissue, detection lanine aminotransferase(ALT),aspartate aminotransferase(AST),triglyceride(TG),totalcholesterol(TC)fasting blood glucose(FBS),free fatty acids(FFAs),tumor necrosis facto alpha (TNF-a) were measured by drawing blood from Abdominal artery; then, the liver tissues were made to 10%liver homogenate for measuring the levels of liver TG,TC, FFAs.Hepatic steatosis was assessed by using H-E stain sections,expression of LXRa of mRNA and SREBP-lc of mRNA in liver tissue was detected by semi-quantitative method(RT-PCR).the expression of liver X receptorα(LXRα) test by immunohistochemical and image analysis.Results:The aspet of fattery libver in model group was typical,but in the lower,middle,higher,positive group recovered in some degree. Compared with normal control,the level of BW was not significantly higher; Compared with untreated model rats,the level of BW was not significantly higher.As compared with the model group,the hygro-weight and liver index of liver of liver lowered,the liver function got better,the level of ALT,AST,TG,TC,FFA also released dramatically.the fat content in liver,eg:TG,TC lowered makably. the level of TNF-αwas not decreased in treated group rats.The fatty degeneration of liver cell and infilomatory cell infiltrion in the model group was moderate to high,but all treating groups had a trend to get better. The expressions of LXRa mRNA,SREBP-1c mRNA and LXRa protein were significantly increased in untreated model rats.The expressions of LXRαmRNA,SREBP-1c mRNA and LXRa protein were significantly decreased in treated group rats.Conclusion:The ultraicro zhikangyin can have the treatment functions to non-alcoholic fatty liver in the model of rats and have protective effects on the liver, can certainly reduce lipid content effect. It also can alleviate lier inflammation mobility, has the good curative effect;can effectively reduce the NAFLD rat glucolipid level, reduce the liver of lipid deposition, has certain treatment on NAFLD.The mechanism may be it could reduce the expression of LXRαand SREBP-1c in fatty liver.
