The phenomena of mixing the false with the genuine and selling shoddy medicines, which is often replaced, has badly restricted the security of employing traditional Chinese medicine (TCMs) and the stabilization of the curative effects, and it has also restricted the developments of the research and application of TCMs. Although the components of TCMs are extremely complex, the rapid development of modern hyphenated instruments and related statistical and mathematical methods make it possible to analyze TCM system and control its quality. The technology of chromatographic fingerprint is widely accepted by international society, in these years, the quality control by chromatographic fingerprint got lots of achievement. Nonlinear chemical fingerprinting(NCF) which is without of pretreat such as separation and purification and suitable for TCMs of various phases and dosage forms, is developing rapidly for its fast analysis and low cost. This thesis aims at the practical TCM system, and carrys out research on chromatographic fingerprint and NCF in order to control its quality and provide useful information for the research of herbal medicine. This thesis includes two parts:the first part of this thesis emphasizes on the applications of high performance liquid chromatography-diode array detector (HPLC-DAD) and correlative similarity comparative analysis(SCA) methods to the research of Radix Codonopsis from 10 different localities and their intermediate materials and end products. The contains include:(1) the selection of suitable extract methods of Radix Codonopsis, optimization of chromatographic conditions and the validation of methodology; (2) comparison and analysis of the similarities of three kinds of processed products by SCA in order to make assessment on the firing process. The results indicate that some components in TCMs are lost or convert to other substance, so investigating the quality and firing technology of TCMs would inclule not only herbal products but also their processed products. The second part is focus on the combinative application of the HPLC fingerprinting and nonlinear chemical fingerprinting to identify the producing areas of Salvia Miltiorrhiza. And the similarity, accuracy and application of NCF are researched in this part. This thesis shows the advantage and huge potential of HPLC fingerprinting in the research of quality control and firing technology of TCMs, and provides a new method on investigating and valuation firing technology. And the thesis illustrates the application of combination chromatographic fingerprint and Nonlinear chemical fingerprinting. This new method gives great help for quality control of TCMs.
