| Purpose:To Study the influence of TangKe decoction of compound Chinese herbs on the positive expression of the apoptosis of isletβcells and apoptosis related protein Fas in type 2 daibetes mellitus rats.Material and method:Rat model of type 2 diabetes mellitus was duplicated by using low-dose strepozotocin(STZ) and high-caloric diet,and using TangKe decoction of compound Chinese herbs and rosiglitazone actes on the model rats individually. By using methods of homeostasis model assessment of isletβcells(HOMA-β),hematoxylin—erosin pigment, TUNEL ,immunohistochemistry and optical microscopic observation,etc, to evaluate the function of HOMA-β, observe the morphology of isletβcell and the positive expression of apoptosis of isletβcells and apoptosis related protein Fas.Results:1.HOMA-β: Before the treatment ,model group was significantly lower than normal control group level in HOMA-β(P<0.01).After the treatment,the levels of HOMA-βof TangKe decoction of compound Chinese herbs group and rosiglitazone group are significantly lower than normal group, but they are all significantly higher than that of model group(P<0.05); And there is no significant difference in the levels of HOMA-βbetween TangKe decoction of compound Chinese herbs group and rosiglitazone group(P>0.05).2.Morphological observation of islets: Islets of model group rats have atrophic shapes ,rough sketch, irregular margins,less cells with sparse-uneven distribution,and empty bubbles can be found. After the treatment,TangKe decoction of compound Chinese herbs group and rosiglitazone group have more cells with dense distribution, and the condition of rosiglitazone group is better than Chinese medicine group. 3.Positive expression of apoptosis of isletβcells: After the treatment, model group show the higher positive expression of apoptosis of isletβcells significantly than normal control group( P<0.01);the positive expression of apoptosis of isletβcells of TangKe decoction of compound Chinese herbs group and rosiglitazone group are significantly weaker than model group(P<0.05),but rosiglitazone group shows a weaker positive expression than TangKe decoction of compound Chinese herbs group(P<0.05). 4.Positive expression of apoptosis related protein Fas: After the treatment, TangKe decoction of compound Chinese herbs group and rosiglitazone group all show weaker positive expressions of Fas in isletβcells than model group obviously (P<0.01), but show superior positive expressions of Fas in isletβcells than normal group in deep extent(P<0.01). It presents striking difference on the positive expressions of Fas in islet cells between TangKe decoction of compound Chinese herbs group and rosiglitazone group(P<0.05),in addition, rosiglitazone group shows weaker positive expressions of Fas than TangKe decoction of compound Chinese herbs group.Conclusion:1.TangKe decoction of compound Chinese herbs can stimulate insulin sectetion of isletβcells and improve the function of isletβcells significantly.2.TangKe decoction of compound Chinese herbs group can obviously increase the number of isletβcells.3.TangKe decoction of compound Chinese herbs can obviously inhibite the positive expression of apoptosis related protein Fas and obviously inhibite the apoptosis of isletβcell as well. |
