Objective:To investigate the effete of salvia miltiorrhiza on NGF,BDNF activity after spinal cord injury (SCI) in rats.Methods:Fifty-five healthy adult SD rats regardless of male or female, weighting 200-250 g, provided by the experimental animal center of Liaoning university of traditional Chinese medicine. Rats were randomly divided into 3 groups: sham operated group simple laminectomy (n=5), pseudo-therapy group (n=25), experimental group (n=25). Pseudo-therapy group and experimental group re-divided into 5 groups (n=5). The SCI animal model was using a modification of the Allen's weight dropping technique. 6 ml/kg of SM was used to the experimental group intraperitoneally and the pseudo-therapy groups were treated with the same volume of saline vehicle 30 min after injury. Each model was given the same dosage once a day until sacrifice or lasted for 5 days following the injury if the rats survived for more than 5 days. At the time points of 1day, 3days, 7days, 14days and 21days following the injury, the rats of two groups which related to the time point were killed to collect the spinal cord tissue samples from the injured segment with the length of l cm. Each sample got histological sections of 10um thickness and were prepared to staining HE and to test the NGF,BDNF by immunohistochemistry method separately. The number of NGF BDNF in every section was counted,and the average positive NGF and BDNF cells of each group at the same time point were calculated. Software SPSS11.5 was applied to analysis if there was significant difference between the two related groups at each time point.Results:The results of HE staining, sham operated group presented the normal morphology neurons and dense white matter. In pseudo-therapy group and experimental group, 1 and 3 days after injury, the gray matter of damage segment appears apparent hemorrhage and a large number of inflammatory cells infiltrate into the damage zone. At 7, 14, 21 day the damage region of spinal cord is reduced in experimental group, and the damage in pseudo-therapy group was severely compared with experimental group. The results of NGF, BDNF immunohistochemistry were positive between pseudo-therapy group and experimental group. There was no significant difference on expression of NGF, BDNF between pseudo-therapy group and experimental group at 1day. The number of NGF and BDNF positive cells were significantly higher in experimental group than pseudo-therapy group at 3, 7, 14, 21d after spinal cord injury(P<0.05).Conclusions:Salvia miltiorrhiza can increase the expressions of the NGF, BDNF of the SD rats after spinal cord injury. It has neuroprotective effects on the second injury mechanism of the spinal cord injury.
