Benzodia.zepines are a class of sedatives and hypnotics, and 33 of them are internationally controlled. At present, more than 10 kinds of these drugs such as triazolam, alprazolam are often used in our country. They are often used for various crimes such as robbery after hypnotizing the victims, which harms people抯 life and property tremendously. These drugs are strongly effective when taken orally at lower doses. Most of them are rapidly metabolized in the body, and the metabolites quickly are excreted out of the body into urine. The concentrations of the metabolites are higher in urine than in blood, and therefore, the qualitative and quantitative analysis of benzodiazepines and their metabolites in urine, which has been an important job for forensic scientists, is of special significance. In this paper, 9 urinary metabolites: a -hydroxytriazolam, a - hydroxyalprazolam, 7-aminoclonazepam, 7-aminoflunitrazepamn, 7- aminonitrazepam, lorazepam, oxazepam, nordiazepam and hydroxyethylflurazepam, which are major metabolites of triazolam, alprazolam, clonazepam, flunitrazepam, nitrazepam, lorazepam, oxazepam, chlordiazepoxide and flurazepam and so on, were measured by GC/MS. Because most of the metabolites contain OH groups or NH groups, the paper mainly developed GC/MS methods for systematic study?on the analysis of the trimethylsilyl(TMS) and t-butyl- dimethylsilyl(TBDMS) derivatives of the metabolites in urine, and the urinary excretion curves of the metabolites of benzodiazepam often encountered were measured. In these reseaches, GC/MS analysis of the TMS derivative of 7-aminonitrazepam. the TBDMS derivatives of 7- aminoclonazepam, 7-aminoflunitrazepam, 7-aminonitrazepam have not been reported at home and abroad. GCIMS analysis of the derivatives of above metabolites in urine except the TMS derivative of a - hydroxytriazolaxn.has not been reported at home In the analysis, the charactoristic mass chromatography was used for qualitative analysis, and the internal standard curves for quantitative analysis. The method is sensitive: LODs were 0.5?ngIml for TMS 2 Abstract ?derivatization, O.2?ngIml for TBDMS derivatization; LOQs were 1?lOng/ml and O.5?ngIml for TMS and TBDMS derivatization, respectively, the repeatabilitiy is good: RSDs were 2.2--9.6% for TMS derivatives and 2.1?.9% for TBDMS derivatives. The accuracy is high: relative recoveries were 93.7?04.3%(TMS derivatization), and 94.O?04.5% (TBDMS derivatization). Linear range was from lOng/mi to 500ng/tnl for each metabolite. The method is of higher specificity: the other ben.zodiazepines and substances which stem from urine did not interfere with the measure of the metabolites.
