Recombinant Human Interleukin-13 Promotes The Dami Cell Proliferation And Expression Of C-mpl Gene

Aim:In this study we investigated the effects of recombinant human interleukin-13(rhIL-13) on the proliferation and expressionof proto-oncogene c-mpl of Dami Cell-a human megakaryobiastic leukemia cell line. methods: The method of growth curve and counting cell are used to investigate the proliferation of Dami cells.The synthesis of DNA in Dami cell is investigated with. methyl. green and pyronin Y cytochemical method. The expression of c-mpl mRNA in Dami Cell is investigated with RT-PCR method.Results: 1. Results of the effect of rhIL-13 on the proliferation of Dami cells: (1). From the growth curve, the numbers and the proliferation rate of Dami cells in rhIL-13 group were far more than that in control group. (2).. The results of counting cell experiment showed the numbers of Dami cells in the rhIL?3 group were 1.41 times as many as that in the control group. significantly different (P<0.01). (3). The results of methyl green-pyronin γ cytochemical method showed that the integral optical density of DNA in rhIL-13 group was 1. 29 times as many as that in control group, significantly different (P<0.01). 2. Results of interaction of rhIL?3 and rhTPO on the growth of Dami2cells (1). From the growth curve, the numbers and the proliferation rate of Dami cells in rhIL-13+rhTPO group were more than that in rhIL-13 group and control group. (2). The results of counting cell experiment showed the rhTPO and rhIL-13 had interaction on the proliferation of Dami cells (P<0.05) (3)The results of orthogonal experimental design showed that the rhTPO. and rhIL-13 had interaction on the synthesis of DNA in Dami cell (P<0. 01) .3. In RT-PCR experiment ,we found the quantitis of expression of c-mpl mRNA in 25ng/ml rhIL-13 group, 50mg/mi rhIL-13 group and 100ng/ml rhIL-13 group increased by 24.8%, 27. 9%, 31.5%.conclusion: Our data indicate that rhILi3 stimulates the proliferation of Dami cells ,moreover, the effects of rhIL-13 on the proliferation are synergetic with rhTPO. In addition, these results suggest that stimulation may be partly by increasing the proto-oncogene c-mpl expression in Dami cells.
