Expression of p21WAFl/CIPl mRNA and protein in bladder transitional cell carcinomasPostgraduate student Wen JinSupervisor Ge GenAbstractObjective To investigate the significance of p21WAFl/CIPl(a cyclin-dependent kinase inhibitor) expression in bladder transitional cell carcinomas(TCC).Method We analyzed p21 mRNA and p21 protein expression of 56 bladder TCC by in situ hybridization and imrmmohistochemical techniques.p21WAFl/CIPl expression was analyzed together with their relation to clinicopathologic parameters of tumors.Result The positive expression rate of p21 mRNA is 51.8%(29/56) and that of p21 protein is 35.7%(20/56).p21 mRNA and p21 protein was negatively correlated with stage and grade of tumors(p<0.05).Conclusion p21WAFl/CIPl is one of the valuable predictors for the biological behavior of bladder TCC and may be a tumor suppressor in bladder carcinogenesis. In situ hybrization and immunohistochemical technique are very useful in evaluating the expression of genes.
