The phenomenon, reperfusion after ischemia of heart for a period of time aggravates the ischemic injury to it ,is called myocardial ischemic-reperfusion (I/R)injury. With the development of cardiovascular surgery, the effects of myocardial protection is becoming more and more important. Heart surgeons have been study how to reduce ischemic-reperfusion (I/R)injury to minium as possible as they can. Up to date, the mechanism of ischemic-reperfusion (I/R)injury has not been expounded completely. But the effect of free radicals(FR)and introcellar calcium overload is widespreadly accepted as the main cause of I/R injury. The hypothesis of FR holds the opinion that I/R injury is caused by free radicals. The effect of antioxidation defensive system(AODS) of human body is to demolish FR. Glutathione peroxidase(GPx) is the main component of AODS. Selenium(Se) is absolutely necessary for maintaining the activity of GPx. It has been testified by a lot of study that supplement of Se can raise the activity of GPx. So, on the basis of theoretic analysis, we can believe that supplement of Se should improve the expressing level of GPx and relieve myocardial I/R injury. Our study is just based on this point and aim to investigate the influence of selenium on the expression of glutathione peroxidase of patients with rheumatismal heart disease.Objective To investigate the influence of selenium on the expression of glutathione peroxidase of patients with rheumatismal heart disease. Methods 30 patients with rheumatismal heart disease ( RHD ) were separated into control group ( 15 patients )and test group ( 15 patients ). Patients in test group received Se supplement each day for two weeks. At the same time, both control group and test group were divided into MVR ( mitral valve replacement ) group and DVR ( mitral valve replacement and aortic valve replacement ) group. Techniques of molecular biology were taken to evaluate and campare GPx expression during I/R injury. Results Both before ischemia and at 30 minutes of myocardial reperfusion, test group had much higher myocardial GPx mRNA expression than control group. And this situation was the same in both MVR group and DVR group. Conclusion The supplement of Se can improve myocardial GPx mRNA expression and defend against free radical superoxidizing damages to the myocardium.So, supplement of Se can improve myocardial protection.
