Biological markers such as mdr1 RNA expression and DNA ploidy status are of important prognostic information in breast cancer. In this study, (1) a fluorescence quantitative RT-PCR detection of mdr1 RNA has been established, with which 38 malignant breast tumor samples and 10 benign breast tumor samples were detected, (2) DNA pliody was determined by flow cytometry in 20 fresh breast cancer samples and 20 paraffin embedded breast cancer samples. The results showed that mdr1 RNA was expressed in 16 out of 38 breast cancer samples (42.11%), among which low mdr1 RNA expression made up 87.5% (14/16), whereas only 1 benign breast tumor samples expressed mdr1 RNA (1/10). In term of DNA ploidy status, 6 (30%) diploid, 1 (5%) tetraploid and 13 (65%) aneuploid were found in 20 fresh breast cancer samples; 4 (20%) diploid, 2 (10%) tetraploid and 14 (70%) aneuploid were detected in 20 paraffin embedded breast cancer samples. Therefore, both methods have been proved to be reliable, reproducible and quick as well, with which important biological markers could be determined in breast tumor.
