IL-6, Il-6r And Study On Relationship Between FAS And Fasl, Bcl-2 And Gastric Cancer

Objectives:The one purpose of the present thesis is to study the expression and regulation of IL-6 and its receptor(IL-6R) in normal gastric mucosas,precancerous lesion,early gastric carcinoma and advanced gastric carcinoma,and to explore if the expression of IL-6 and IL-6R are related to the differentiated degree of gastric carcinoma and the metastasis of lymph nodes,and to discuss their clinical significance,and to provide a theoretic basis for early diagnosis and immune treatment of gastric carcinoma.The other purpose of the present thesis is to study the expression and regulation of Fas,FasL and Bcl-2 in normal gastric mucosas,precancerous lesion.well-moderate differentiated gastric carcinoma and poorly differentiated gastric carcinoma,and to provide a theoretic basis for the diagnosis and genetic treatment of gastric carcinoma. MethodstThe expression of IL-6 and IL-6R mRNA were detected by means of In situ hybridization(ISH) in 20 cases of normal mucosas,20 cases of chronic atrophic gastritis(precancerous lesion) ,25 cases of early gastric carcinoma and 26 cases of advanced gastric carcinoma.And the expression of Fas,FasL and Bcl-2 at protein level were detected by means of immunohistochemical technique in 20 cases of normal mucosas,23cases of chronic atrophic gastritis,25 cases of well-moderate differentiated gastric carcinoma and poorly differentiated gastric carcinoma.Result:The positive expression rates of IL-6 mRNA were 10%,55%,84% and 84.6% in normal mucosas,precancerous lesion,early gastric carcinoma and advanced gastric carcinoma,and those of IL-6R mRNA were 10%,50%,80% and88.4% respectively.The positive rates of IL-6 and IL-6R were significantly different between normal gastric mucosas and precancerous lesion(P<0.05),and there were significant difference for the expression of IL-6 and IL-6R between precancerous lesion and early gastric carcinoma(P<0.05),whereas no statistical difference were found for expression of IL-6 and IL-6R between early gastric carcinoma and advanced gastric carcinoma, the expression of IL-6 and its receptor were not related to the differentiated degree of gastric carcinoma cell and the metastasis of lymph nodes.But there were obvious relationship between the expression of IL-6 and the expression of IL-6R in the different lesions of gastric mucosas.The positive expression rates of Fas were 95% for normal mucosas,91.3% for precancerous lesion,60% for well-moderate differentiated gastric carcinoma and 40% for poorly differentiated gastric carcinoma,respectively.The levels of Fas expression in gastric carcinoma were obviously lower than those in both normal mucosas and precancerous lesion (P<0.05),whereas significant difference were found neither between normal mucosas and precancerous lesion,nor between well-moderate differentiated gastric carcinoma and poorly gastric carcinoma.The positive expression rates of FasL were 25%,56.5%,88.2% and 84%,respectively.Significant difference were found both between normal mucosas and precancerous lesion and between precancerous lesion and gastric carcinoma,whereas there was obvious difference between well-moderate differentiated gastric carcinoma and poorly differentiated gastric carcinoma.The positive expression rates of Bcl-2 were 20%,78.3%,80% and 64%,respectively.There was significant difference for expression of Bcl-2between normal gastric mucosas and both precancerous and gastric carcinoma,whereas no obvious difference was found among precancerous lesion,well-moderate differentiated gastric carcinoma and poorly differentiated gastric carcinoma.Conclusions:In the clinicopathological process of normal gastric mucosas,precancerous lesion,early gastric carcinoma and advanced gastric carcinoma,the positive expression rates of IL-6 and IL-6R increase gradually.Especially,the positive expression of IL-6 and its receptor are found in precancerous lesion and early gastric carcinoma.and there is obvious relationship between the expression of IL-6 and IL-6R.But the expression of IL-6 and IL-6R are not related to the d...
