Objective: To investigate the expression of NHERF1protein in normalgastric mucosa and gastric carcinoma and its relationship with expression ofCD44v6and MMP-7, Meanwhile detect the expression of NHERF1mRNA innormal gastric mucosa and gastric carcinoma. Furtherly to study the correlationbetween NHERF1gene expression and tumorgenesis, development andbiological behaviour in gastric carcinoma.Methods:1.All cases(paraffin embedding specimens) were from department ofpathology of xingtai people’s hospital in October2009to October2011. Atotal of48cases of gastric cancer group, aged39-78, including29cases ofhigh-middle differentiation adenocarcinoma, poorly differentiatedadenocarcinoma19cases; Male34cases, female14cases; Lymph nodemetastasis positive group of32cases,16cases of negative group.Allspecimens have not been preoperative radiotherapy and chemotherapy, at thesame time take10cases of gastric mucosa of distance from tumor tissue>5cm as normal controls.2.The expressions of NHERF1, CD44v6and MMP-7protein in48cases ofgastric carcinoma and10cases of normal gastric tissues were detected byimmunohistochemistry.3.The expression of NHERF1mRNA in48cases of gastric carcinoma, and10cases of normal gastric tissues was detected by in situ hybridization.4.Statistical Product and Services Solutions(SPSS) statisticalsoftware(version11.5) was applied for data analysis.x2test,Speraman bankrelation analysis were used,with P<0.05for a statistically difference.Results:1The expression of NHERF1gene 1.1The expression of NHERF1protein: The positive expression rate ofNHERF1protein in gastric carcinoma and normal gastric mucosa was33.3%、90.0%resepectively, and there was statistical difference among the2groups(χ2=8.649,P<0.05). The expression of NHERF1protein was related to thediffertiated degree of gastric carcinoma, depth of invasion and lymph nodemetastasis (P<0.05), but it was not correlated with gender and tumordiameter(P>0.05).1.2The expression of NHERF1mRNA: The positive expression rate ofNHERF1mRNA in gastric carcinoma was lower than that of normal gastricmucosa. The expression of NHERF1mRNA was related to the differtiateddegree of gastric carcinoma, depth of invasion and lymph node metastasis(P<0.05), but it was not correlated with gender and tumor diameter(P>0.05).2The expression of CD44v6proteinThe positive expression rate of CD44v6protein in gastric carcinoma washigher than that of normal gastric mucosa. The expression of CD44v6proteinwas related to depth of invasion and lymph node metastasis (P<0.05), but itwas not correlated with differtiated degree of gastric carcinoma, gender andtumor diameter(P>0.05).3The expression of MMP-7proteinThe positive expression rate of MMP-7protein in gastric carcinoma andnormal gastric mucosa was70.8%、20.0%resepectively, and there wasstatistical difference among the2groups (χ2=7.052,P<0.05).The expression ofMMP-7protein was related to depth of invasion and lymph node metastasis(P<0.05), but it was not correlated with differtiated degree of gastric carcinoma,gender and tumor diameter(P>0.05).4In gastric carcinoma the expression of NHERF1protein was negativelycorrelated with those of CD44v6、 MMP-7protein (r=-0.399,P<0.05;r=-0.421,P<0.05), but the expression of CD44v6protein was positivelycorrelated with that of MMP-7protein(r=0.348,P<0.05). The expression ofNHERF1mRNA was positively correlated with NHERF1protein in gastriccarcinoma (r=0.907, P<0.05). Conclusion:1. The positive rate of NHERF1expression in gastric carcinoma is remarklylower than that in normal gastric tissues, the positive rate of CD44v6andMMP-7expression in gastric carcinoma is remarkly higher than that in normalgastric tissues. It suggest that low expression of NHERF1and high expressionof CD44v6, MMP-7may be associated with tumorgenesis and developmentin gastric carcinoma.2. The expression of NHERF1, CD44v6and MMP-7proteins were related todepth of invasion and lymph node metastasis. It suggest that low expression ofNHERF1and high expression of CD44v6, MMP-7may be associated withinvasion and metastasis in gastric carcinoma.3. The identical decreased expressions both NHERF1protein and mRNAindicate that the expression of NHERF1gene is inhibited at the level oftranscription or before.
