The Study Of Relationship Between Survivin Protein And Ovarian Neoplasm

Object:(1)To investigate the expression of Survivin protein in epithelial ovarian neoplasm;(2)To explore the expression difference of Survivin in human ovarian cancer cell line SKOV3 treated with different concentrations of topotecon(TPT);(3)To investigate the apoptosis rate and the invasion and metastases ability of human ovarian cancer cell line SKOV3 treated with different concentrations of topotecon(TPT).Methods:(1)Survivin protein was detected by immunohistochemical technique and tissue chip method in 20 cases of normal ovarian tissues,25 cases of benign tumors,9 cases of ovarian borderline tumors and 54 cases of ovarian cancers.The relationship between the expression of Survivin and clinic data of ovarian carcinoma was analyzed;(2) Immunohistochemistry was used to measure the expression of Survivin protein in human ovarian cancer cell line SKOV3;(3)The FCM(Flow Cytometry)was used to measure apoptosis rate of human ovarian cancer cell line SKOV3 treated with different concentrations of topotecon (TPT);(4)semi-quantitative gelatin zymography was used to measure the secretion of MMP-2,9 by SKOV3 cells,Transwell chamber assay was performed to determine its effect on invasion and migration capacities of SKOV3 cells.Results:(1)The Survivin expression in malignant ovarian tumor and ovarian borderline tumor is significantly higher than benign ovarian tumor and normal ovaries(P＜0.007),However there was no significant difference between malignant ovarian tumor and ovarian borderline tumor(P=0.267),between benign tumor and normal ovarian tissue(P=0.117).Survivin expression was related with clinic stages,the positive rate of Survivin in stageⅢ-Ⅳ(78.4%) was significantly higher than that in stageⅠ-Ⅱ(41.2%)(P=0.007).The positive rate of Survivin was increased with the tumor differentiation degrees decreasing(P=0.027).The Survivin positive rate had a significant difference between those with abdominal dropsy(94.7%)and those without abdominal dropsy(51.4%)(P＜0.001),between those with lymph node metastasis(96.7%)and those without lymph node metastasis(29.2%)(P＜0.001).However there was no significant difference between serous cystadenocarcinoma,mucinous cystadenocarcinoma and other types of ovarian cancer(P=0.977);(2)Survivin was expressed in SKOV3,and positive rate in SKOV3 was decreased with the increasing of TPT concentration,There was a significant difference between each groups with different concentrations of TPT(P＜0.01);(3)There were a significant difference about the cell apoptosis rate between each groups with different consistency of TPT(P＜0.05);(4)There were a significant difference about the secretion of MMP-2,9 between each groups with different consistency of TPT(P＜0.05),and as the same result of the rate of invasion of each group(P＜0.05).Conclusion:(1)The expression of Survivin is correlated with malignant behavior of ovarian tumor,the more malignant,the higher expression rate of Survivin,Survivin may play an important role in carcinogenesis and development of ovarian cancer,and it may be valuable to further understand the biological behavior of ovarian carcinoma.;(2)TPT can inhibit the expression of Survivin;(3)Survivin is an important factor related with apoptosis,invasion and metastases ability;(4)TPT can inhibit the proliferation and induce the apoptosis of SKOV3 cell lines and decrease the cell invasion and metastases ability of SKOV3 cell lines in vitro.
