The secondary reperfusion injury caused by various reasons followed cerebral ischemia have important influence on the prognosis and treatment of ischemic stroke. This paper focus on the inflammation damage in reperfusion injury, and divided into two parts, namely the document research and exTKrimental research..The document research include: inflammation cytokine, adhesive molecular and cerebral ischemia damage, traditional Chinese medicine aimed to therapy of cerebral ischemia/reperfusion damage.In the experimental research part, we use the method of thread-occluding to make reversible middle cerebral artery occlusion and reperfusion (MCAOR) in rats.甇bserving the content of inflammation cytokine (TNF- aand IL-1P)> intercellular adhesive molecular-1 and von Willebrand factor in brain tissue of rats suffered ischemia 2h followed by reperfusion 2h> 6h> 12h. 24h, 48h, 72h> 120hu 168h to find the expression pattern of TNF- a, IL-lp> ICAM-1 and vWF, which will instruct the selecting of time point for drug application (D observing the influence of cholalic acid, jasminoidin and their compatibility on the expression of TNF-cu IL-lp, ICAM-L vWFandNSE.The results showed:1. Expression pattern of TNF- a in brain tissueIn rats underwent middle cerebral artery occlusion (MCAO) for 2h followed by 2h reperfusion, the content of TNF- a protein showed evident increase, and peaked at reperfusion 6h and 120h, content of TNF- a protein at reperfusion 120h was higher than that of reperfusion 6h.The high level of TNF- a protein sustained to reperfusion 168h(p<0.01).The results of immunohistochemistry showed that the sham-operated group had a weak expression of TNF- a .The immunoreactive cells are neurons and glial cells. At reperfusion 24h and 72h,the model group demonstrated a marked increase of TNF- a in neurons and glial cells, the number of immunoreactive cell and expression intensity at reperfusion 72h were higher than those of reperfusion 24h.2. Expression pattern of IL-1 Pin brain tissueIn rats underwent MCAOR, the content of IL-1 P protein significantly increased at reperfusion 6h(compared to sham-operated group, pO.Ol), sustained to reperfusion 48h and decreased at 72h, the content of IL-1 P protein from reperfusion 72h to 168h returned to almost same level as sham-operated group.3. Expression pattern of ICAM-1 in brain tissueCompared to sham-operated group, ICAM-1 protein significantly increased during reperfusion 6h to 48h ( p<0.05 or pO.Ol ), and the content of ICAM-1 protein from reperfusion 72h to 168h returned to almost same level as sham-operated group.As judged by immunohistochemistry, the sham-operated group had a weak expression of ICAM-1 , few ICAM-1 positive maicrovascular endothelial cells were observed By reperfusion 24h,-5-Ihe expression intensity of ICAM-1 and the number of ICAM-1 positive vessels were significantly increased, and the leukocyte adhesived to microvascular endothelial cells also had the expression of ICAM-1. The number of ICAM-1 positive vessels and expression intensity at reperfusion 72h were higher than those of reperfusion 24h.The content of ICAM-1 protein during its expression course had significant correlation with that of IL-1 P protein, which suggest that IL-1 P probably is the initiating factor of ICAM-1 expressioa The content of IL-1 P protein during its expression course also had significant correlation with that of TNF-cc protein, and because TNF- a 's expression is ahead of IL-1 P 's expression, so maybe TNF-ot could stimulate the expression of IL-1 P . 4. The content of vWFin plasma during different reperfusion time pointThe content of vWF in plasma significantly increased in rats underwent MCAO for 2h followed by 6h reperfusion, and sustained to reperfusion 24h, The content of vWF in plasma at reperfusion 48h had no significant statistic difference with that of sham-operated group.
