This paper includes two parts: documental research and experimental research.The documental research includes two parts: fundamental research of cascade of damage of cerebral ischemia, traditional Chinese medicine aimed to the treatment of cascade of damage of cerebral ischemia and reperfusion injury.The experimental research includes two parts: in the first part, adult male SD rats received Oh,0.5h,lh3i,6h,12h24h,48h,72h,120h of permanent middle cerebral artery occlusion(MCAO) with an intraluminal suture technique. Measuring the level of brain superoxide dismutase (SOD) and Malondialdehyde (MDA) and the level of tumor necrosis factor - alpha (TNF- a ), interlukin - Ibeta (IL-1 (3 X intercelluar adhesive molecular-1 (ICAM-1) in brain to observe their relationship with cascade of damage of cerebral ischemia Measuring the level of vonWillebrand factor (vWF) in rat's plasma and Neuron-specific enolase (NSE) in rat's serum to observe the condition of endothelial cells and neurons during cascade of damage of cerebral ischemia The second part, observing the influence of Cholalic acid, Jasminoiclin and their compatibility on the expression of SOD , MDA, TNF-cu IL-lp, ICAM-1 , vWF and NSE to discuss the protection of Chinese herb (Cholalic acid, Jasminoidin and their compatibility) on the cascade of damage of cerebral ischemiaThe results showed:The expression condition of SOD , MDA, TNF-a, IL-lp, ICAM-1, vWFandNSE:1. Malondialdehyde (MDA) began to increase 0.5h after MCAO, peaked at 0.5h to lh,and reached a second peak at 48h to 72h,and persisted to 120hat the same time, vitality of superoxide dismutase (SOD) began to decrease 0.5h after MCAO, and the decrease persisted to 120h2. In rats underwent MCAO for lh,the content of TNF- aprotein showed evident increase, and peaked at this time, the high level of TNF- aprotein sustained to 168h(p<0.01). The results of irnrnunohistochemistry showed that the sham-operated group had a weak expression of TNF- a, the immunoreactive cell are neurons and glial cells. At MCAO for 24h and 72h,the model group demonstrated a marked increase of TNF- a in neurons and glial cells, the immunoreactive cell are mainly microglial cells, at 72h,endothelial cells had a expression of TNF- a too. And TNF- a expression intensity at 72h was higher than those of 24hThe content of IL-1 P protein began to increase 0.5h after MCAO, from 0.5h to 168h,tne content of IL-1 (3 protein maintains a high level compared to the sham-operated group.Compared to sham-operated group, ICAM-1 protein significantiy increased during 12h to 72h after MCAO.As judged by immurwhistochernistry, the sham-operated group had a weak expression of ICAM-1, few ICAM-1 positive vascular endothelial cells were observed, by 24h and 72h after MCAO, the expression intensity of ICAM-1 of endothelial cells and the number of ICAM-1-positive vessels were significantly increased-7-3. The content of vWF in plasma began increased in rats underwent MCAO for lh,peaked at 24h to48h^ndpersistedtol20haflerMCAO.The content ofNSE in serum began increased in rats underwent MCAO for 0.5h, The content ofNSEinserijmsignfficantIyinaeasedat6h^ndpeakedat48h, and 1he increase persisted to 120h.The results of immunohistochernistry showed that neurons of sham-operated group had a specific expression of NSE, the high density immunoreactive cells were regularly distributed and the tainting were well-distributed, while 24h and 72h after MCAO, the density of immunoreactive cells was evidently decreased, the cells scattered irregularly, and manifested a weak expression on the whole.4. The result of HE staining showed that the brain tissues of sham-operated group are normal. While 24h and 72h after MCAO, neurons of the cortex of model group manifested evident loss and the density of them decreased. The condensed cytoplasmic of neurons showed red dyeing and nuclei of neurons showed Shrinkage.
