P27kip1 With Hematologic Malignancies

ObjectiveTo evaluate the expression of cyclin dependent kinase inhibitor P27(Kipl) and to investigate its clinical significance in acute non-lymphocytic leukemia (ANLL), acute lymphocytic leukemia (ALL), chronic myeloid leukemia (CML) and chronic lymphocytic leukemia (CLL).MethodsThe P27(Kipl) protein was measured by Western blot and enhanced chemoluminenscence (ECL) on bone marrow or peripheral blood samples in 82 cases of leukemia.Results1. The expression of P27(Kipl) protein in ALL is higher than that in ANLL (P=0.033) and also higher than that in CML (P=0.008). P27(Kipl) expression in CLL is higher than that in CML too (P=0.017).2. In acute leukemia, the effective rate (CR and PR) of initial chemical therapy was higher in the group of P27(Kipl)>0.655 than that in the group of P27(Kipl) P=0.655, P=0.041.3. In ANLL and ALL patients, the survival time was longer in the group of P27(Kipl)>0.655 than that in the group of P27(Kipl) < 0.655, P=0.0065. There were similar statistical significance in ANLL patients and ALL patients, respectively (P=0.0271 and P=0.0266).4. The expression of P27(Kipl) protein appeared no relationship with patients' chromosomal abnormalities, sex, age, white blood cell number or blast cell number in peripheral blood, serum LDH or serum uric acid.5. Compared with Tripure ?method and normal cell lysing method, therewas no significant difference in protein extracting efficiency. It was important to improve test sensitivity that 6 X sample loading buffer,narrow well and thick gel in electrophoresis was used, protein was transferred to PVDF membrane with 80 voltage isobarically for 1.5hours, and X-ray film was exposed as soon as chemiluminescence reaction was finished. ConclusionsIn acute leukemia, lower P27(Kipl) expression might associated with poor prognosis.
