Objective The effects of crocin on Cell proliferation capacity and cytokine release of stimulated peripheral blood mononuclear cells by phytohemagglutinin (PHA) were examined.Methods The monomuclesr cells were isolated from peripheral blood of children with acute lymphocytic leukemia for complete remission by the methods of Ficoll-Hypaque and divided into one control group、phytohemagglutinin (PHA) group and four experimental groups(A-D),which propotional crocin final concentration were0.625,1.25,2.5and5mg/ml. After3days culturation,the effects of four concentrations of crocin, phytohemagglutinin, and phosphate buffered solution on cell proliferation and production of cytokines, including interleukin (IL)-2and IL-4were evaluated by MTT assay, the IL-2ELISA kit and the IL-4ELISA kit.Using the flow cytometry,the T lymphocyte subset distribution were detected.Result In cells stimulated with PHA, different concentrations (from1.25mg/ml to5mg/ml). of crocin significantly promoted cell proliferation of lymphocytes and the proliferation index gradually increased when the crocin concentration increased,the difference among them were significant(P<0.05). High concentrations of the crocin(2.5and5mg/mL,respectively) also more promoted secretion of IL-2in stimulated cells than that of PHA,and the production of IL-2was the highest when the concentration of crocin was5mg/ml.(P<0.05). The effects of crocin at graded dose levels from1.25to5mg/ml on IL-4secretion were higher than that of PHA,and IL-4also increase the concentration when crocin increased concentration.(P<0.05). The ratios of IL-2to IL-4in the presence of all concentrations of saffron and PHA on stimulated cells were significantly higher than for the control group (P<0.01). There is a higher CD4/CD8ratio of T subset in the concentration of crocin for5mg/ml than that in control group.(P<0.05).Conclusion The crocin showed a stimulatory effect on IL-2and IL-4secretion and cell proliferation in stimulated cells by PHA.①The crocin can promote lymphocyte proliferation,and present dosage dependence;the proliferation index was gradually increased within the concentration1.25-5mg/ml.②The production of IL-2was higher when the crocin concentration was2.5-5mg/ml than that of PHA and IL-2concentration is the highest when crocin was5mg/ml.③IL-4secretion was gradually enhanced compared to that of PHA group when crocin concentration was1.25、2.5and5mg/ml.
