Bifidobacterium represent a numerically significant group in the adult human and animal colon, and members of this genus have previously been shown to have positive effects on the health of host. Potential benefits include alleviation of lactose intolerance, ambition of pathogenic micro - organisms and viruses, reduction of cholesterol levels , tumor inhibitory effects, activation of the immune system, and production of votaries.The intestinal intraepithelial lymphocytes (iIEL) , which have a very strong cytotixicity, at as an important part of the intestinal musical immunity. The ilEL have the killing activity through mitogen - induced cytotoxicity and Ab - dependent cell mediate cytotoxicity. Otherwise , the ilEL can secrete a lot of cytokines , such as IL - 2, IL -3,IL-4, IL-5, IL-6, IFN7, TNFα and TGFβ. By secreting IL-2, ilEL can stimulate NK cells and enhance their ability of killing tumor cells. Because IL -2 is a paracellular cytokine, it can stimulate the synthesis of local IgA . The new research indicate that the IFN secreted by ilEL participate the infections immunity of Anti - HIV and vi-nis, In vitro, ilEL can secrete some cytokines to enhance the antigen present of intestinal mouse. Taken together, ilEL participate the intestinal musical immunity by severed ways, to execute its functions of an-ti - infection and anti - tumor.The experiment judged the applicative value of bifidobacterim at the base of ilEL function. We extracted high purified ilEL from the intestinal and identified their antigen phenotype. The ilEL inoculated directly with ceU and extinction of bifidobacteion and E. codi, andtested the natural killing activity, the production of IL - 2, IFNγ of ilEL. So we can judge the applicative value of bifidobacterial solvents and investigable its mechanism, which will provide the theories evidence of application.First, we extracted the high purified iIEL by mechanic separation and non - constant density gradient centrifugation, and tested their antigen phenotype by directed immunofluorescence, at the same time, we extract the polysaccharide of bifidobacteria and E. coli by organic solvents. After than use had them co - manipulated, and fest the ability of natural killing and the production of IFNγ and IL - 12 in iIEL. Results; (1) The phenotype of iIEL:85. 1\2.8% , CD3 + cell, 73.8 \2.1% , CD8+ cell, Expression of TCR 43. 8\2.4% ; (2) The a-bility of natural killing in bifidobacteria is 53. 75\5. 21% , in E. coli is 32.6\3. 54% , which in control is 16. 92\1. 98% . So there is significant difference between Bifidobacteria, E. coli and the control. The bifidobacteria is more strong. (4) The production of IFNγ: 166. 5\4.7u/ml in bifidobacteria; 55.9 \3. lu/ml in E. coli; 27.4 \2. 8u/ml in control, so there is significant difference. Bifidobacteria and E. col enhance the ability of natural killing of iIEL, and promote the ilEL to produce the IFNγ and IL -2, but the effect of bifidobacteria is stronger .The antigen phenotype of cellular membrane in ilEL, express more than 73. 8% CD8 + T cell and 43. 8% TCRγδ, which is corresponding to the references. The references suggest that there are 40. 6% TCRapT cell and 46.7% TCR^cell in ilEL cells. Because there are few l6T cell in other tissues, ilEL B the repertoire of^Tcell. The large emergence of 8 T cell in intestinal makes people to pay attention to its function mucosal immunity mucous and epithelial fissure is thefirst defense of body ageist microbes, and they are also the area where the tumors occur. So the ilEL has the prominant state in and - man-ropes , anti - parasites, and anti - tuner. Now, ilEL are found to have significant cytotoxicity, which can lysis the target cells directly bike cryonic t cells ( CTL, CDg + ) ; ilEL can produce many cytosine, like CD4 + Th cell.
