Studies on the Effects of polygona-polysaccharose's upon APPtransgenic mices' learning and memory ability and its functionAlzheimer's disease(AD)is one of the most widely seen diseases ofdementia. It is characterized by the damage of memory and cognition. Apatient who suffers from dementia will lose the capacity of looking afterhimself, thus aggravate the burden of the whole family and the society.With the appearance of more and more aged-people, the incidence of ADwill grow continuously. Therefore, to investigate AD basically andclinically has greatly clinic and social significance.The cause of AD is unknown till now, but most researchers think thatan excessive amount of Aβinduces neuron degeneration and apoptosis bydepositing in the brain. This might be the main reason of the patient'sdecrease of cognition. Polygona-polysaccharose is an effective drug topostpone aging and increase immunity. Therefore, mice which had beentransferred genes of APP were studied in this experiment. The aim of thepresent study is to investigate the effects of polygona-polysaccharose onthe spatial learning and memory of APP transgenic mice, the influence ofpolygona-polysaccharose on biochemical indexes and hippocampusstructure in APP transgenic mice, the expression of amyloid-β-protein (Aβ)and the expression of choline acetyltransferase (CHAT) in APP transgenic mice and the influence of polygona-polysaccharose on them. The purposeis to explore the effects and functions of polygona-polysaccharose on APPtransgenic mice, and try to find a new way of preventing and treatingexperimental AD.Objective: To investigate the effect of polygona-polysaccharose onAPP transgenic mice, and to study the protective and therapeutic functionof polygona-polysaccharose on APP transgenic mice.Method: Inject different concentration of polygona-polysaccharosesolution into the mices' stomach continuously for 45 days. The Morriswater maze was adopted to study the ability of the mices' initiative learningand memory; light microscope was used to observe morphology; theexpression of Aβand ChAT was detected by immunochemistry tissuestaining method. Eventually, the effect of polygona-polysaccharose on theabove-mentioned index was investigated.Result: The experimental result shows that polygona-polysaccharosehas the following effects:1. Ethology: compared with the treat group, the delitescence of theBlank group prolonged obviously, the frequency of finding the platformdecreased, and the frequency of mistake increased (p<0.01). Within thetreat group, the group with high concentration polygona-polysaccharoseincreased in the frequency of finding the platform, shortened in thedelitescence, and decreased in the frequency of mistake (p<0.05). 2. Expression of ChAT and Aβ: compared with the Blank group, theenergy of ChAT in the treat group was heightened, and the expression ofAβwas degraded (p<0.01). Within the treat group, there is no difference.3. Morphology change: the amount of neuron of the blank was lessthan that of the treat group. Cell Apoptosis phenomenon could be seen, forexample, cell nucleus of some neuron showed pyknosis condition,caryotheca slightly thickened and aberration lipofuscin increased and so on.Polygona-polysaccharose treat group had more neurons, the changes of cellwere not obvious, cell had normal structure.Conclusion: Polygona-polysaccharose can improve the ability ofleaming and memory of APP transgenic mice. The protective andtherapeutic mechanism of polygona-polysaccharose can protect the centralcholinergic system and resist Aβdepositing. Polygona-polysaccharose is aneffective prescription for the prevention and therapy of experimental AD.
